Abstract:To explore the transmission characters of chromosomes from Thinopyrum elongatum in the background of durum, The lines Du_No.2 and Du_No.4 derived from a cross between Tritileytrigia 8801(AABBEE) and T. turgidum L. ssp. durum(AABB) were characterized by chromosome-specific molecular markers, genomic in situ hybridization(GISH), and non-denaturing fluorescence in situ hybridization(ND-FISH). The results of molecular markers indicated that lines Du_No.2 and Du_No.4 could respectively amplify the specific band of 2E,4E chromosome from Th. elongatum. The results of GISH and ND-FISH showed that lines Du_No.2 and Du_No.4 had a 2E and 4E chromosomes of Th. elongatum respectively. The results of meiosis indicated that both lines have E chromosome segregation abnormalities in anaphase I, II and telophase II of meiosis. In addition, the positive and negative crosses between the two lines and durum displayed that chromosomes 2E and 4E could be transmitted through male gametes in durum background, but could not be transmitted through female gametes. This study provided a basis for the establishment of a complete set of durum-Th. elongatum addition lines and substitution lines.
