Morphological analysis and molecular identification of filamentous cyanobacteria from Jinyang Lake

    摘要:

    蓝藻是地球上最古老的生物之一,其形态结构较为简单,为产氧型光合作用的原核生物。目前对于淡水湖泊丝状蓝藻的多样性研究相对较少。本研究从山西省晋阳湖分离、纯化出5株丝状蓝藻藻种通过显微镜直接观察显微结构(如细胞形状、藻丝体宽度、是否有鞘),大致为三个科(Leptolyngbyaceae、Pseudanabaenaceae、Desertifilaceae),进一步以16S rRNA基因为标记,构建系统发育树,进行分子鉴定。结果表明:分离藻株的形态学鉴定和16S rRNA序列系统发育分析结果相一致。其中2株为Leptolyngbyaceae(JYH005和JYH012),2株为Pseudanabaenaceae(JYH008和JYH022),1株丝状蓝藻为Desertifilaceae(JYH010)。

    Abstract:

    Cyanobacteria is one of the oldest organisms on earth. It is prokaryotes of oxygen-producing photosynthesis with relatively simple morphology and structure. Until now, there are relatively few studies on the diversity of cyanobacteria in fresh-water lakes. In this study, five filamentous cyanobacteria species were isolated and purified from Jinyang Lake, Shanxi Province. The microstructure (such as cell shape, size and sheath postural) were directly observed under the microscope, which was roughly divided into three families (Leptolyngbyaceae, Pseudoanabaenaceae and Desertlaceae). Further, the phylogenetic tree was constructed with 16S rRNA gene as marker for molecular identification. The results showed that the morphological identification of the isolated algae strains was consistent with the results of 16S rRNA sequence phylogenetic analysis. Among them, 2 strains were leptolyngbyaceae (JYH005 and JYH012), 2 strains were pseudoanabaenaceae (JYH008 and JYH022), and 1 strain of filamentous cyanobacteria was desirtiferaceae (JYH010).
