    UV-B对植物生长具有重要影响,UVR8是目前植物中发现唯一接收UV-B的光信号受体。为研究甘薯紫外光受体UVR8的功能,采用CODEHOP结合RACE技术,设计兼并引物,克隆得到UVR8全长cDNA序列(IbUVR8)。IbUVR8 (NCBI登录号KT749986)的ORF全长为1329bp,共编码了441个氨基酸,相对分子量为47745.20,理论等电点pI为5.72。IbUVR8与番茄、马铃薯UVR8蛋白序列相似性高,进化相对保守。RT-qPCR结果显示,IbUVR8在甘薯中具有组织特异性,表达量为叶片>茎>根。在低温、干旱和NaCl等非生物胁迫处理下, 紫、绿、黄三种叶色甘薯的IbUVR8除了绿叶甘薯(福菜薯18号)在NaCl胁迫下表现为先上调后下降外,其余均为上调表达。三种叶色甘薯在GA3处理下表现为微弱上调,同时在Cu2+处理下则表达差异显著。推测该基因可以响应非生物胁迫和激素信号的诱导。原核表达获得大小与预期一致的IbUVR8-GST融合蛋白。

    Abstract:

    UV-B had an important effect on plant growth. UVR8 was the only light signal receptor found in plants to receive UV-B. In order to understand function of sweet potato UVR8, we used CODEHOP combined with RACE technology to clone the full length of UVR8 cDNA sequence (IbUVR8). The ORF of IbUVR8 (NCBI accession number KT749986) had a full length of 1329 bp, encoded a total of 441 amino acids. IbUVR8 had a relative molecular weight of 47745.20, and a theoretical isoelectric point pI of 5.72. IbUVR8 had high similarity with tomato and potato UVR8 protein sequence, and UVR8 evolution was relatively conservative. RT-qPCR results showed that IbUVR8 had tissue specificity in sweet potato, and the expression level was leaf>stem>root. In abiotic stress, IbUVR8 was up-regulated under three leaf-colored sweet potato varieties under low temperature, drought and NaCl stress. Fucaishu 18 was up-regulated first and then decreased under NaCl stress. The three leaf-color sweet potato varieties showed a slight up-regulation under GA3 stress. Under the Cu2+ treatment, the expression of three leaf-color sweet potato varieties were significantly different. It is speculated that IbUVR8 can respond to the induction of abiotic stress and hormone signals. Prokaryotic expression obtained IbUVR8-GST fusion protein with the same size as expected.
