    WUSCHEL-related homeobox (WOX)转录因子是植物特有的转录因子,具有调控植物的生长发育和非生物胁迫响应的作用。本研究通过生物信息学的方法对甘草WOX基因家族进行序列分析,并通过实时荧光定量PCR技术(RT-qPCR)检测该基因家族在干旱、盐、无磷、脱落酸(ABA)和赤霉素(GA3)处理下的表达量。结果显示,(1)甘草WOX基因家族共有16个成员(GuWOX1~GuWOX16),其氨基酸数目在168~452 aa,蛋白分子量在19.39~50.11 kDa,等电点分布在5.28~8.97,均含有保守的HD结构域,为亲水性不稳定蛋白质。(2)系统进化树分析表明WOX基因家族分为3个进化支(远古支、中间支和现代支)。(3)启动子序列中包括响应多种逆境和激素的顺式作用元件。(4)RT-qPCR结果显示,在不同时间段下,GuWOXs在盐、无磷和GA处理下大部分被诱导表达,干旱处理和ABA处理的表达模式相近,在处理12 h时均上调表达。研究表明,WOX转录因子在甘草逆境胁迫中起调控作用,该研究结果为进一步分析WOX基因家族成员在甘草抗逆境胁迫等方面奠定基础。

    Abstract:

    WUSCHEL-related homeobox (WOX) transcription factors are plant-specific transcription factors that can regulate plant growth and development and the response to abiotic stress. In this study, the sequence of Glycyrrhiza uralensis WOX gene family was analyzed by bioinformatics, and the expression of this gene family under drought, salt, phosphorus free, abscisic acid (ABA) and gibberellin a3 (GA3) treatment were detected by real-time fluorescence quantitative PCR (RT-qPCR). The results showed that: (1) there were 16 members of the G. uralensis WOX family (GuWOX1-GuWOX16). The amino acids encoded by WOX genes were variable from 168 to 452 aa,with the protein molecular weight ranged from 19.39 to 50.11 KDa and The theoretical isoelectric point distribution ranged from 5.28 to 8.97. GuWOX contained conserved HD domains and were hydrophilic and unstable proteins. (2) Phylogenetic tree analysis showed that the WOX family was divided into 3 major clades, (e.g., the Ancient clade, the Intermediate clade and the Modern/WUS clade). (3) Numerous stress and hormone-responsive cis-elements were observed in the GuWOX promoter regions. (4)RT-qPCR analysis showed that most GuWOXs were induced under salt, phosphorus-free and GA treatments at different time periods. The expression patterns of GuWOXs under drought and ABA treatments were similar, and the expressions of GuWOXs were up-regulated after 12 h of treatment. This indicates that GuWOX transcription factor played a regulatory role in stress of G. uralensis. The results provided a foundation for further analysis of WOX gene family members in stress resistance of G. uralensis.
