Abstract:
In order to explore a new approach to rapid propagation of Quercus acutissima Carr.,the somatic embryos were induced from the mature embryos.And then the somatic embryos were cultured into seedlings.The results revealed that the best medium to induce the somatic embryos was MS+1.0 mg·L-1 6-BA+1.0 mg·L-1 IBA+1.0 g·L-1 L-glutamine+0.5 g·L-1 proline on which the induction rate could reach to 70.0% in 30 d.The optimal mature medium was 1/2MS+2.0 mg·L-1 6-BA+0.5 mg·L-1 IBA+2.0 mg·L-1 ABA+4.0 g·L-1 L-glutamine+2.0 g·L-1 proline on which most of somatic embryos were fully mature in 60 d.The best germination medium was 1/2MS+0.2 mg·L-1 6-BA+10 g·L-1 sorbitol,and cold treatment was conducive for germination.The germination rate could reach to 100.0%.The regenerated plants formed in 80 d.
