    Abstract:

    Laser confocal scanning microscope (LCSM) and whole-cell patch clamp were used to reveal the influence of actin cytoskeleton deploymerizing agent cytochalasin B (CB)and stabile reagent phalloidin (PD) on the dynamic of cytosolic free calcium concentration ([Ca2+]i) and calcium ion channels in the protoplasm membrane of Pyrus pyrifolia pollen tube cutting-edge.The results showed that CB could promote [Ca2+]i increase and activate Ca2+ channel in plasma membrane;while phalloidin had no effect on [Ca2+]i and did not activate Ca2+ channel.The results indicated that disruption of actin cytoskeleton might inhibit pollen germination and tube growth by activating Ca2+ channel in plasma membrane and increasing the concentration of [Ca2+]i.
