Abstract:长豇豆花药壁的发育为基本型。绒毡层周原质团型,小孢子母细胞减数分裂粗线期至末期Ⅱ,相邻小孢子母细胞间存在胞质通道。四分体多为四面体型,少数为左右对称型。生殖细胞刚形成时,壁呈PAS负反应,以后为PAS正反应。生殖细胞游离在营养细胞质内后,壁消失。成熟花粉为2细胞型。在小孢子早期,首次观察到在周原质团型绒毡层细胞外切向壁的外方,有很多直径约2-4μm,含不溶性多糖、蛋白质和抗乙酰解物质的球形颗粒,以后这些颗粒沿绒毡层细胞的径向壁进入药室,同时在小孢子壁和原生质体之间也出现了同样的颗粒。小孢子外壁增厚的同时,颗粒消失。关于这些颗粒的作用,有待进一步深入探讨。

Abstract:过去所得的实验表明,植物中ATPase活性变化与耐冷性有关。春小麦和番茄对低温是非常敏感的,当将这两种植物的幼苗在低温下培养时,质膜ATPase活性下降甚至消失。相反,在冬小麦中,当幼苗在低温下处理时,质膜ATPase活性增加.为了研究ATPase同工酶变化与植物耐冷性之间的关系,以小麦等四种植物为材料,在0-1℃低温下处理2天、2周和4周,然后检测幼苗根系内的ATPase同工酶,结果发现,在0-1℃培养时,幼苗根系内ATPase同工酶谱带减少,ATPase同工酶谱带变化与植物幼苗耐冷性呈一定相关性。ATPase同工酶谱带变化可能是受冷害所致。

Abstract:对5种基因型幼胚诱导的愈伤组织继代培养表明,玉米胚性愈伤组织的长期继代受基因型,培养基成分,激素,培养条件的影响。适时继代,逐代筛选对胚性保持起重要作用。适当降低培养温度(12±2℃)有利于愈伤组织的保存和胚性保持,可以减少愈伤组织长期继代所需的物质和工作量。长期继代培养的胚性愈伤组织,胚状体发生能力和植株再生率无显著变化,但正常苗的再生频率显著下降。观察愈伤组织细胞染色体发现:(1)基因型对不同倍性细胞的比例有明显影响。(2)随着继代时间的延长,二倍体细胞下降,四倍体和非二倍体细胞增多。(3)愈伤组织中出现多种染色体结构变异,这些结构变异有可能导致非整倍体细胞的形成。
