Abstract:利用在禾谷类作物中表达效率较高的启动子Ubi对含目的基因sgna的现有载体进行了改造,并引入筛选标记基因bar;为提高目的基因的表达水平,在目的基因5′端引入了Ω和kozak序列,3′端引入了poly(A)序列,成功构建了适用于小麦的抗虫基因植物表达载体pGU4AGBar和pGBIU4AGBar,基因pGU4AGBar含有顺向连接的Ubi-sgna及Ubi-bar基因表达盒,pGBIU4AGBar含有T-DNA边界序列,并在其左右边界中间插入了含有顺向连接的CaNV35S-nptⅡ,Ubi-sgna和Ubi-bar基因表达盒,人工合成的雪花莲外源凝集素基因sgna可以编码对同翅目昆虫具有毒杀作用的蛋白。

Abstract:用带有ACC脱氨酶基因和卡那霉素抗性基因(NptⅡ,作为报告基因)的工程根癌农杆菌转化白兰瓜子叶。通过组织培养,得到具有卡那霉素(Km)抗性的再生小苗,经NptⅡ报告基因的PCR扩增,ACC脱氨酶基因的Southern点杂交以及ACC脱氨酶活性的生理生化检测可知,ACC脱氨酶基因已成功转入白兰瓜子叶再生小苗,而且不同植株的真叶中表现出不同水平的ACC脱氨酶活性。

Abstract:利用RT-PCR的方法,从豇豆种子,子叶及成熟叶片中分别提取RNA,反转录后以该基因两端的保守序列为引物进行PCR扩增,PCR产物与pUCm-T载体连接进行克隆,蓝白筛选重组子,经测序,其核苷酸同源性高达100%,蛋白质同源性达91%。本试验同时对该基因的表达时期差异进行了大致检测。结果是种子的表达量最高,子叶其次,成熟叶中最低。

Abstract:在数量性状水平上,调查了43个春小麦品种的19个数量性状,利用主成分分析法计算品种间的欧氏平方遗传距离。并在此基础上用最小方差法做了聚类分析。发现43个品种间在数量性状水平上的遗传距离变异范围很大(0.926-67.942)。平均遗传距离为18.000,说明供试品种的19个考察性状上存在较大的表型变异,从聚类结果来看,地方品种基本上被聚在一起,说明地方品种同引进品种和育成品种至少在表型上存在较大差异。从20世纪40年代以来,在数量水平上甘,青两省春小麦地方品种间的遗传多样性水平最高(GD=31.389),其次是20世纪50年代引进品种(GD=26.308),而育成品种间的遗传多样性水平最低,总体上呈下降趋势,说明随着育种进程的深入,作为育种目标追求的经济性状趋于一致,其变异集中在一个狭小的范围之内。品种间的遗传多样性下降。

Abstract:研究了保水剂对干旱胁迫下刺槐叶绿素a荧光动力学参数的影响。结果表明,在干旱胁迫下刺槐叶绿素a荧光动力学参数Fv/Fm,δPSⅡ,qp,NPQ均有所下降,且随着保水剂用量增大,下降程度减小,即保水剂用量越大,对受刺槐干旱胁迫的缓解作用愈明显。保水剂不同水平处理对刺槐叶绿素荧光参数的影响达到显著水平。多重比较结果表明,土壤中保水剂用量只有在不低于0.5kg/m^3时才能有效地发挥抗旱作用。

Abstract:在深达50-60cm的有效土层中,研究了矮化砧M26不同利用方式对富士苹果树根群分布及生育的影响。结果表明:(1)M26作中间砧用时因栽植深度不同,富士苹果树的根鲜重,干重均增加,树势健壮,T/R也高;(2)M26作砧木用的长度不同,富士苹果树地下,地上部生长各异,砧木微乎其微三的根鲜重,干重比砧木长度长的增加,T/R率下降,但矮化效果不明显;(3)根群的水平分布,栽植深度深的主要集中在距主干80-100cm的范围内,栽植浅的集中在距主干60-80cm范围内,砧木长度长的主要集中在50-60cm范围内,砧木长度短的主要集中在70-80cm范围内,两种方式的根群水平分布行间大于株间;(4)根群垂直分布主要集中在0-60cm的土层中,占根系总量的96%以上,根群密集区分布于20-40cm的士层中,占总根量的70%以上。

Abstract:在河北保定生态条件下,研究了京、冀,鲁,豫等北方小麦主产省份近年来育成的15个小麦推广品种的16种品质性状和产量表现。结果表明;这些品种可分为2大类,第1类为优质型品种,加工品质表现较好,其中豫麦34表现既优质又高产,中优9507具有突出的优良品质特性;第2类品种为高产型品种,产量高但品质性状较差,不同性状具有各自的遗传变异特点,仍需要通过各种途径扩大和丰富我国小麦育种材料品质和产量性状的变异性,沉降值作为重要品质性状,遗传力较高,与大多数面粉,面团品质参数显著相关,在育种早代选择中应予重视,基于所有品种的相关分析得出产量与沉降值,粉质仪测定参数存在负相关,但以10个优质品种为材料的分析结果表明,产量与品质性状以及品质必状间的不利相关被打破,使对生产有利的性状得到了协调与结合改良。
