Abstract:采用磷酸铅沉淀的电镜细胞化学方法,对经冷驯化及未经冷驯化的金边卫矛 EuonymusradicansEmorald&Gold. 以及两者在-5℃下处理1d后的叶肉细胞进行了ATPase的超微细胞化学定位的比较观察,并对冷驯化可提高植物抗寒力及ATPase与植物抗寒性关系进行了探讨.标志ATPase活性反应的磷酸铅沉淀物主要分布在叶片栅栏细胞和海绵细胞的质膜和液泡膜上,ATPase在这两类细胞中的分布及活性无明显差异.金边卫矛幼苗经4℃低温驯化7d后,其质膜和液泡膜的ATPase活性高于22℃下生长的幼苗,冷驯化后的金边卫矛幼苗在-5℃处理1d后,其质膜和液泡膜上仍可观察到ATPase的活性反应,但较低温胁迫前有所降低,且超微结构完整,未见任何伤害.而未经冷驯化的金边卫矛幼苗在-5℃处理1d后,其质膜和液泡膜上的ATPase完全失活,同时细胞的超微结构受到严重伤害.实验结果表明,冷驯化可提高金边卫矛幼苗ATPase在低温胁迫下的稳定性,这种活性变化与植物抗寒力的提高呈正相关,同时还发现液泡膜ATPase活性较质膜ATPase变化幅度大,推测液泡膜可能较质膜ATPase对低温更为敏感.

Abstract:利用基因工程技术构建了含有几丁质酶(Chitinase,Chi.)、β-1,3-葡聚糖酶(β-1,3-Glucanase,Glu.)、萝卜抗真菌蛋白(Raphanus sativus-antifingal protein,Rs-AFP2)的三价植物表达载体,并通过农杆菌直接转化法将三价植物表达载体转入农杆菌EHA105,分别为GC R/pCAMBIA1300/EHA105和RC G/pCAMBIAl300/EHA105,并采用PCR、DNA dot blotting技术对其进行了鉴定分析,证明获得了阳性的重组农杆菌工程菌株。

Abstract:盐胁迫下,DK961(耐盐)和LM15(盐敏感)小麦种子的发芽率(Gr)、发芽指数(Gi)和活力指数(Vi)均显著下降,LM15下降的幅度大于DK961.外源100 mg/L GA3、1×10-7 mol/L ABA和0.1% Ca(NO3)2处理均能缓解盐对小麦种子萌发的抑制作用,对盐胁迫下LM15种子萌发的缓解作用显著好于DK961,并且不同程度地缓解盐处理引起的种子内源GA 1+3含量和α淀粉酶的活性下降,从而降低盐胁迫对种子萌发的抑制作用.表明盐抑制小麦种子萌发的主要原因是盐胁迫导致种子内源GA 1+3含量和α淀粉酶的活性下降.

Abstract:通过对澳洲青苹茎尖培养及植株再生体系中无菌培养物的建立、初代培养、继代培养、生根培养和移栽五个技术环节的研究,建立了澳洲青苹组织培养快速繁殖技术体系。结果表明:(1)BA0.3~0.6mg/L NAA0.1~0.3mg/L的MS培养基能促进澳洲青苹茎尖生长或大量侧芽的分化,不定芽生长健壮;(2)无根幼苗在含有IBA0.6~1.0mg/L NAA0.1mg/L的1/2MS培养基上暗培养一周后,再进行自然光培养,30d生根率可达到78%以上;(3)生根苗经50mg/L NAA浸渍1~2h后,移入蛭石:腐殖土:田园土=2:1:1的移栽基质中,保温保湿,成活率达80%以上;(4)进一步建立了自然光下生根、练苗一次完成的同步化体系,缩短了繁育时间,简化了步骤,降低了成本。此技术体系为澳洲青苹优良种苗快速繁殖提供了一条新途径。

Abstract:对农业废弃物无害化处理后,添加一定的无机肥后制作成有机无机复混肥,使其肥料化资源利用.为了进一步研究有机无机复混肥的肥效,2001~2003年在夏玉米上进行了连续3年田间试验,试验结果表明,有机无机复混肥可以促进玉米生长,增加玉米生物产量1958.0~2770.5kg·hm-2,改善玉米产量结构,提高玉米籽粒产量866.0~1144.5kg·hm-2.并能降低玉米耗水系数,提高玉米水分利用效率2.680~3.399kg/ mm·hm2 .在旱地施用有机无机复混肥,可以提高旱地水资源的利用效率.

Abstract:采用HPLC法对陕西省冬油菜区5个试区种植的双低油菜籽粒中硫苷总量和组分含量进行了分析,结果表明: 1 陕西省不同地区的生态条件对双低油菜总硫苷含量有显著的影响,相差达13.14μmol/g,CV为13.30%,5个试区硫苷含量的排序为宝鸡>安康>咸阳>大荔>汉中,地区间达显著或极显著差异;品种间硫苷含量的稳定性有明显的差异,以P01-10品种稳定性最好; 2 在测定的8个硫苷组分中,变幅最大的5个组分依次为2-羟基-3-丁烯基硫苷、4-羟基-3吲哚甲基硫苷、4-戊烯基硫苷、3-丁烯基硫苷和2羟基-4-戊烯基硫苷,极差分别为:5.70、2.69、1.70、1.31和1.03μmol/g,变异系数分别为:17.59%、16.41%、58.84%、10.21%和50.32%,其中以3-丁烯基硫苷稳定性最好,4-戊烯基硫苷稳定性最差; 3 籽粒中硫苷与含油量、千粒重及容重均有显著的相关性,相关系数分别为-0.783、-0.948和0.656,通径和偏相关分析表明,仅硫苷与千粒重偏相关显著为-0.859,含油量和容重是通过千粒重来影响硫苷的.
