Abstract:对农杆菌介导法转化棉花技术体系进行了综合改进,突破基因型的限制,使多品种或基因型(10个以上)的我国主栽棉花品种(系)转化成功,其中中棉所24号发展成为快速模式化转化品种(转化周期5~7个月,转化率8.1%),CCRI27、CCRI36等多个品种转化体系基本成熟。不同转化体系的愈伤组织诱导率为10%~40%,愈伤组织胚性愈伤诱导率达到10%~40%,转化率总体平均达到1.8%,并对大批量再生苗的鉴定予以程序化。实现棉花转基因规模化,达到年产转基因棉花植株2000~4000株以上的水平。

Abstract:以5~7d龄的 丽春 番茄无菌苗子叶作为外植体,研究了子叶外植体对抗生素卡那霉素的敏感性,抗生素卡那霉素对番茄筛选的适宜浓度为70mg/L.通过根癌农杆菌 Agrobacteriumtumefaciens 介导,成功地进行了马槟榔甜蛋白基因MBLII对番茄的遗传转化,获得转化番茄抗性植株,组织化学法有阳性表现、PCR特异扩增及Southern杂交检测出现特异条带,表明MBLII基因已顺利整合到转基因番茄植株的基因组.

Abstract:在人工气候室水培条件下,选用3个不同抗旱类型的冬小麦品种,研究了水分胁迫对冬小麦幼苗叶绿素荧光参数的影响。结果表明:水分胁迫下,冬小麦幼苗可变荧光(Fv)、最大荧光(Fm)、可变荧光与最大荧光比(Fv/Fm)、可变荧光与初始荧光比(Fv/Fo)、光化学淬灭系数(qF)均降低;而初始荧光(Fo)与非光化学淬灭系数(qNP)则升高,说明光系统Ⅱ(PSⅡ)受到了伤害,使得PSⅡ原初光能转换效率(Fv/Fm)、PSⅡ潜在活性(Fv/Fo)降低;光合电子传递、光合原初反应过程受到抑制,起光保护作用的热耗散提高。但水分胁迫下品种间各参数变化幅度不同。除qNF外,其余各参数均为抗旱性越强降低幅度越小,而qNF则升高幅度越大。说明水分胁迫对冬小麦幼苗叶绿素荧光参数的影响与其抗旱性密切相关。

Abstract:在CO2浓度倍增(700 μmol·mol-1)条件下,以5个不同剂量的UV-B辐射对大棚番茄的光合作用及SOD、POD、CAT酶活性的影响进行了研究.结果表明CO2倍增能够明显提高番茄叶绿素含量、净光合速率和抗氧化酶活性.在CO2倍增条件下,低剂量UV-B辐射(<1.163 kJ·m-2·d-1)可以刺激番茄叶片叶绿素含量升高,抗氧化酶活性升高,与CO2的正效应有叠加现象,但对光合作用的影响不大;高剂量的UV-B(>1.163 kJ·m-2·d-1)辐射使植株的叶绿素含量、净光合速率和抗氧化酶活性降低,对植物产生胁迫作用,CO2倍增与UV-B辐射复合处理可以减弱和部分抵消这种抑制作用.

Abstract:研究了蛋白核小球藻在增强UV—B辐射下NO信号的产生。结果表明:NO释放量与UV—B的强度相关。在藻类细胞培养液中加入NOS的竞争性抑制剂硝基精氨酸(LNNA),不能抑制NO的释放。加入NO清除剂乙酰半胱氨酸(NAC),能明显减少NO的释放。在无氮和有氮培养基中NO同样的释放。这些结果说明NO生成途径不经过依赖L-精氨酸的NOS和依赖NO3^-和硝酸还原酶,NO的底物不是L-精氨酸和NO3^-在无氮和有氮培养基中同样有NO3^-的形成,NAC减少NO2^-的释放,说明生成了NO2^-有可能是NO释放后生成的。

Abstract:模拟兰州地区夏季臭氧层减薄12%和20%时增强的UV-B辐射2.54kJ·m-2·d-1 T1 和4.25kJ·m-2·d-1 T2 ,以自然辐射为对照,研究2种辐射对番茄幼苗生长、叶绿素和可溶性蛋白质含量以及过氧化物酶 POD 、吲哚乙酸氧化酶 IAA氧化酶 活性的影响.结果表明:低强度、短时间的辐射 T1,10d 对番茄幼苗生长没有影响,而高强度、长时间的辐射可减少生物量累积,降低株高、壮苗指数、叶绿素a、b和总叶绿素的含量,对叶绿素a/b无显著影响;与对照相比,可溶性蛋白质含量在辐射处理10d和20d时均减低,40d时升高;T2处理使POD活性一直显著高于对照;IAA氧化酶活性在辐射10d和20d时增加,40d时显著低于对照.说明在UV-B辐射下光合下降、IAA分解抑制了番茄幼苗的生长.

Abstract:以4~5叶的 二苠 茄幼苗为试材,研究了其在低温弱光 10℃/5℃昼/夜,光强60、120μmol·m-2·s-1 胁迫7d并恢复7d后的光合特性变化.结果表明,低温弱光胁迫后茄子幼苗的净光合速率、气孔导度和叶绿素含量显著降低;光补偿点、光饱和点、光饱和时的Pn、表观量子产额降低;CO2补偿点升高,CO2饱和点、CO2饱和时的Pn、光合能力、CO2羧化效率降低;以低温下较强光照时 120μmol·m-2·s-1 的变化幅度较大;恢复7d后各项指标仍然不能恢复到对照水平.试验条件已对茄子幼苗叶片光合机构的结构和活性造成了不可恢复的伤害.

Abstract:武陵山地区含有中国种子植物特有属72个,占该区种子植物总属数的7.16%,占全国同类属数的28.69%,占华中同类属数的77.42%。其基本特征为:(1)古老与孑遗的木本植物众多,并存在一定相对年轻的成分;(2)地理成分多样,具明显的亚热带性质,表现出四方汇集和东、西、南、北的过渡性质;(3)水平分布上呈现南、北两个分布中心,垂直分布上以1000m以下的中、低海拔为主;(4)武陵山地区植物区系与神农架、秦岭、湘黔桂交界地区的关系比较密切,而与南岭、武夷山、滇黔桂地区的关系比较疏远。根据武陵山地区中国种子植物特有属的数量、与鄂西~川东特有现象中心(或者三峡中心)所含属的一致性以及地理间的密切联系,可以认为该区是鄂西~川东中心(或者三峡中心)的重要组成部分甚至是核心地带,在华中植物区系中发挥着举足轻重的作用。
