Abstract:以植物的胚珠和子房为实验材料,介绍一种用Steedman's wax 包埋对组织切片中的细胞核进行DAPI染色的方法.Steedman's wax 作为一种低熔点多酯蜡,具有与石蜡相似的性质,切片方法同常规石蜡切片,适合于切成厚度大于5 μm的连续切片.Steedman's wax包埋的切片能成功地进行DAPI染色.与用压片法和Technovit 7100或GMA包埋切片法进行的DAPI染色相比,用Steedman's wax 包埋切片法进行的DAPI染色具有廉价、操作简便、可进行连续切片、图象清晰等优点,特别在植物细胞程序化死亡(PCD)的研究中及细胞核DNA含量测定方面,有着较大的应用价值和潜能.

Abstract:针对翅果油树愈伤组织诱导的褐变问题,从外植体、碳源、抗氧剂的类型及浓度等的筛选进行了研究。结果表明;幼龄的茎段是诱导愈伤组织的最佳材料。葡萄糖为碳源的培养基能有效地防止褐变。抗氧化剂Vc和Na2S2O3能减轻褐变程度,其中0.15g/L的Vc效果最佳,其次为0.1g/L和0.2g/L。Na2S2O3对褐变的抑制作用不及Vc,3种浓度无明显区别。吸附剂AC不能防止外植体褐变,且抑制愈伤组织的诱导。

Abstract:利用体视显微镜、扫描电镜和光学显微镜,对珍稀濒危植物翅果油树(Elaeagnus mollis Diels)的茎、叶、果、休眠芽等多种器官表面覆盖的表皮毛,进行了详细观察.发现其表皮毛的形态和结构具有多样性,各器官有差异,据此将它们分为分支状毛、星状毛、盾状-星状毛、盾状毛四类,以及一些特殊的表皮毛,如:分支状毛与星状毛或盾状-星状毛的过渡类型称为类星状毛和类盾状-星状毛.叶片上表面的表皮毛,为分枝状表皮毛,且稀疏散布.叶片下表面的白色表皮毛,也多为分枝状表皮毛,但分布较密,叶主脉处最为稠密多为星状毛.叶柄的表皮毛形态呈现一定的梯度变化,远轴端(靠近叶片端)为类星状毛,近轴端(靠近茎的一端)为盾状-星状表皮毛,中间段为星状毛介于前二者之间.茎表面灰色的或褐色的表皮毛,以盾状表皮毛为主,夹杂少量盾状-星状表皮毛,因茎的老嫩其表皮毛形态略有不同.外果皮沟槽内的表皮毛多为盾状表皮毛,嵴部则密集星状或分支状表皮毛.休眠芽的鳞片多覆盖盾状表皮毛,只在鳞片的尖端有少量星状毛和分枝状毛.翅果油树多种器官表面多种表皮毛的存在是其适应高山(海拔800~1300m)环境的一种形态特征.表皮毛具有反射阳光、阻止水分过度散失、保温、防止机械损伤等功能.为探究翅果油树耐早、耐寒、耐高温机理以及组织培养过程中外植体易污染等问题,提供了形态学依据,也为进一步进行表皮毛的发育和分子生物学研究打下了一定的基础。

Abstract:以库拉索芦荟、木立芦荟和皂质芦荟为材料,用扫描电镜观察其叶表皮气孔和角质膜的结构,用高效液相色谱法(HPLC)测定了3种芦荟属植物叶中芦荟素的含量。扫描电镜观察结果表明,3种芦荟叶表皮都覆盖有厚的角质膜,气孔下陷,表现出典型的旱生植物特征。但角质膜的纹饰和厚度在不同芦荟间有显着差异。木立芦荟角质膜表面呈瘤状突起,角质膜厚度为5~6μm,库拉索芦荟和皂质芦荟的角质膜表面较平,库拉索芦荟的角质膜厚度为3~4μm,皂质芦荟的角质膜厚度为8~10μm。高效液相色谱法(HPLC)测定结果表明,木立芦荟叶含芦荟素最高,库拉索芦荟叶含量较低,而皂质芦荟叶含量最低。此外,本文还初步探讨了芦荟属植物叶表皮结构与芦荟素含量的关系。

Abstract:在野外实地观测锦带花(Weigela florida(Bunge)A.DC.)的开花动态和访花者,在显微镜下观测柱头和花粉的形态及花粉-胚珠比,用TTC法测定锦带花花粉的活力,用联苯胺-过氧化氢法测定柱头可授性。结果表明,锦带花的花粉活力在开花时达70%以上,且能维持3d,之后明显下降。锦带花的柱头在开花3h后多数呈白色、二裂、具可授性,柱头的可授期持续2~3d。柱头与花药之间有5.3士0.9mm的空间距离,自然条件下难于自花授粉.开花当天7.00~10.00之间访花者最丰富,开花第2天访花者减少,开花第3天时访花者更少。在锦带花上记录到的访花者隶属于膜翅目、鳞翅目和双翅目,其中,熊蜂和蜜蜂是主要传粉者。
