Abstract:佛手是芳香科枸橼类植物,有很好的药用和观赏价值,但其品种较为单一,对其进行遗传改良和品种选育显得十分必要.为了将基因枪转化方法应用于佛手的遗传改良和品种选育,本实验以GUS基因为报告基因,利用基因枪方法将其转入金华佛手外植体内,通过检测GUS基因在佛手叶盘中的瞬时表达情况,分析了外植体的幼嫩程度、射程、氦气压力、真空度、轰击次数等参数对GUS瞬时表达的影响,并研究了最佳检测时间.结果表明,以幼叶为受体,在射程为6 cm、氦气压力为7.584×106Pa,真空度为88.046 kPa条件下,2 d后可检测出GUS基因的最佳表达活性.轰击次数多少对GUS基因瞬时表达影响不明显.

Abstract:用RAPD技术进行新疆核桃早实特性的分子标记研究,用180个10-mer随机引物分别扩增早实和晚实近等基因池DNA,筛选出5个多态性引物,结果只有引物OPG15(5’-ACT GGG ACT C-3’)在早实近等基因池及其个体中能重复扩增出一条约710bp的特异片段OPG15 710,而晚实近等基因池及其个体中无此特异片段,将OPG15 710克隆于pUCm-T载体,筛选阳性克隆用限制性内切酶Pst1消化,表明克隆片段大小正确。实步分析认为,OPG15 710可能是与核桃早实特性相关的分子标记,该标记仍在研究之中,该研究将为进一步克隆早实基因,实现早实优质,探索早实机制奠定基础。

Abstract:根据苎麻组织自身含有较多的本分类,粘状物及黄酮类等次生代谢物质的特点,对CTAB法进行优化后用于提取苎麻组织总DNA。在研磨过程中加入抗氧化剂PVP,提取缓冲液加入终浓度6%的PVP和4%的β-巯基乙醇,可防止整个提取过程中溶液变褐;在第2次用氯仿/异戊醇抽提时,加入10%的CTAB和4%的NaCl高盐溶液,以除去多糖。经几种改良方案提取总DNA的ISSR-PCR扩增结果验证,以同时加入抗氧化剂和高盐溶液的CTAB-4法效果最好。

Abstract:为了探讨低剂量微波对增强UV-B辐射损伤菘蓝(Isatis indigotica Fort.)的修复作用,将经过10.08 kJ·m-2·d-1辐射损伤(PAR=220 μmol·m-2·s-1)的菘蓝幼苗分别经0、3、6、9和12 s等不同时间的微波辐照(126 mW·cm-2,2 450 MHz),然后测定其幼苗MDA含量、紫外吸收物质含量、抗坏血酸含量以及3种抗氧化酶SOD、CAT和POD活性.结果表明,增强UV-B辐射损伤菘蓝在微波的作用下其菘蓝幼苗中SOD、CAT和POD活性及紫外吸收物质含量、抗坏血酸含量提高,MDA含量得到显著的降低,说明微波对增强UV-B辐射伤害菘蓝幼苗具有修复作用.但是,随着微波剂量的增加,这种修复效应减弱,甚至消失.上述参数的变化说明适量时间的微波处理可以提高菘蓝对增强UV-B辐射的抵抗能力,并在此基础上初步探讨了微波的修复机理.

Abstract:采用L I-COR 6400便携式光合测定系统测定并研究了乌兰布和沙漠白刺叶不同水分状况下光合速率及其叶绿素荧光参数的变化特征.结果表明,随着白刺水势降低其净光合速率和气孔导度快速下降,当水势降低到一定值时,净光合速率、气孔导度几乎不变;而叶绿素荧光指标与叶水势关系在高水势时随着叶水势下降叶绿素荧光指标值近似不变,当水势下降到一定值时近似呈直线下降.白刺水饱和状况下的光补偿点为(43.1±4.8)μm o l.m-2.-s 1,光饱和点为(676±150)μm o l.m-2.-s 1;叶生长初期和叶成熟期净光合速率水势补偿点(净光合速率为0时的水势)分别为-3.65 M Pa和-5.76 M Pa,NPQ初始水分胁迫水势分别为-2.20 M Pa和-6.63 M Pa.研究指出运用净光合速率水势补偿点和非光化学猝灭初始水分胁迫水势可评价白刺对干旱环境的适应性.

Abstract:用不同浓度(0、0.1、0.2、0.4 m o l.L-1)的N aC l处理BG 110培养的发菜细胞,结果显示,发菜光合速率与叶绿素荧光强度随N aC l浓度的升高先增加后降低,当N aC l浓度为0.1 m o l.L-1时光合速率与叶绿素荧光具有最大值,表明发菜细胞培养物能耐受一定浓度的盐胁迫.以BG 110+0.4 m o l.L-1N aC l为对照,在BG 11+0.4m o l.L-1N aC l的胁迫实验中,光合速率与叶绿素荧光强度下降较慢;丙二醛、脯氨酸含量较低;类胡萝卜素含量较高,表明在培养液中添加外源硝酸盐后可以缓解N aC l对发菜细胞培养物的生理胁迫效应,增强其抗盐性.

Abstract:运用开顶式气室(OTC-1)研究了O3胁迫对原位条件下冬小麦叶片的光合色素、脂质过氧化和抗氧化系统的影响.结果表明:随着O3浓度的升高,各个生育期小麦叶片中叶绿素含量下降,叶绿素组成发生改变,且在灌浆期变化明显;叶片相对电导率(REC)增大,丙二醛(M DA)含量增加;过氧化氢酶(CAT)、过氧化物酶(POD)活性和类胡萝卜素(C ar)含量降低,超氧化物歧化酶(SOD)活性在O3浓度低于1×10-7(φO3)时逐渐升高,而后急剧降低.可见O3促进了小麦叶片老化,加剧了膜脂过氧化,破坏了抗氧化系统功能,影响了叶片的正常生理代谢.

Abstract:在温室中通过严格控水的方法,对4个不同水分处理下,12个黑杨杂交无性系间长期水分利用效率(WUEL)、δ^13C和光合参数(Gs、Tr、Pn、WUEL,Cj、Pmax、LSP、Ce)的差异进行了研究,并分析了无性系间WUEL和δ^13C差异与光合参数差异的相互关系。结果表明:不同水分处理下,δ^13C和Gs、Tr、Pn、WUEL、Cj差异极显著。随水分胁迫的加剧,δ^13C和WUEL增加,而Gs、Tr、Pn、Cj减少。同一水分处理下无性系间WUEL和δ^13C差异显著,Gs、Tr、Pn、WUEj、Cj差异极显著。无性系问WUEL和δ^13C的差异在4个水分处理下表现出一致性.即WUEL最优的是J2,其次为J6、J7、J8、J9,同时这些无性系的δ^13C较高。Gs是影响WUEj的关键因子,Gs低的无性系WUEj高。J2、J6、J7、J8、J9的净光合速率(Ps)和瞬时水分利用效率(WUEj)虽不是最高,但其最大净光合速率(Pmax)大,光饱和点(LSP)高,羧化效率(Ce)大,光合能力强。无性系间δ^13C和WUEL差异的主要影响因子是Gs和光合能力,J2、J6、J7、J8、J9都具有Gs低和光合能力强的特点。各水分处理下δ^13C与WUEL,均呈正相关,但由于水分引起的δ^13C差异远大于无性系间的差异,因此二者的相关性仅在充分供水条件下表现较好.相关系数为0.7077。

Abstract:本试验对91(260)3-3-8对白粉病的抗性再次进行鉴定,进一步确定该材料对白粉病完全免疫。为了研究其抗白粉病基因的遗传规律,用感病材料陕225、小偃6、邯6172、豫麦49与该材料杂交得到F1、F2后代群体,发现F1抗白粉病;陕225/91(260)3-3-8F2、小偃6/91(260)3-3-8 F2、邯6172/91(260)3-3—8 F2群体抗感比例为3:1,表明“91(260)3—3—8”与感病材料陕225、小偃6、邯6172相比有1对抗白粉病基因的差异;豫麦49/91(260)3-3—8 F2群体抗感比例不完全符合3;1,其原因有待于进一步探明。
