Abstract:以逆境诱导型启动子rd29B为驱动,分别构建了含有抗逆调节转录因子DREB1B基因的表达载体pBAC123、pBAC128,选择标记为bar基因.用高压氦气基因枪PDS1000/He分别将表达载体和p35SIH3导入多年生黑麦草(Lolium perenne)品种Topgun成熟胚的愈伤组织.经除草剂Bialaphos抗性筛选和植株再生,获得了62株转基因植株.经PCR、Dot-blotting分子检测,DREB1B基因已整合到多年生黑麦草部分转基因株系的基因组中.用5种不同浓度的除草剂涂抹黑麦草叶片,非转基因植株表现为不抗,而转基因植株最高可以抗135~200 mg/L.脯氨酸含量测定表明,使用15%PEG8000处理后,转基因植株的叶片脯氨酸含量比非转基因植株提高1倍左右.经25 d人工温室干旱处理,有5棵转基因植株存活;复水后,有3棵植株恢复正常生长.结果表明,利用逆境诱导型启动子(rd29B)来增强外源DREB1B基因的表达,能显著改良黑麦草的抗旱能力.

Abstract:将微卫星探针5′端生物素化后与链亲和素磁珠特异结合,用磁珠和探针的结合物与两端连接已知序列人工接头的中国李品种小黄李(Prunus salicinacv.Xiaohuangli)基因组DNA酶切片段杂交,以此杂交片段为模板用人工接头序列为引物进行PCR扩增,根据PCR产物测序结果设计引物作为微卫星DNA的标记引物.结果在随机挑选的36个克隆进行菌落PCR检测时,从31个阳性克隆中挑选18个克隆进行测序后获得了12条特异序列,设计的8对SSR引物均在5个中国李受试品种上获得了预期的扩增产物,其中4对引物在受试品种上表现出多态性.

Abstract:以白皮松成熟胚为外植体诱导再生小植株.试验结果表明,成熟胚不定芽诱导以MS培养基最佳,附加0.294 mg/L的NAA和3.56 mg/L的6-BA时,诱导率接近100%;MS培养基附加NAA(0.05 mg/L)时,平均增殖系数可达6.3以上.不定芽增殖率最大值(10)出现在SH培养基上,此时NAA浓度为0.05 mg/L、6-BA浓度为4 mg/L.MS培养基中加入适量活性炭和GA3能促进不定芽生长,随着活性炭和GA3浓度的增加,有效嫩梢(≥2cm和≥4 cm)的比率显著增高;当活性炭和GA3浓度过高时(分别超过2.75 g/L和4.1 mg/L),不定芽的伸长与生长受到抑制.在离体培养条件下,以种胚为外植体获得了无根苗.

Abstract:采用溶液培养法对Cd2 胁迫下绿豆和箭舌豌豆幼苗叶组织内的抗氧化酶活性、活性氧和丙二醛(MDA)含量以及细胞电解质泄漏率的变化进行了研究。结果显示,随着Cd2 胁迫浓度的增加和胁迫时间的延长,2种作物叶组织内的MDA含量和细胞电解质泄漏率明显增加;在胁迫期间,二者的过氧化氢(H2O2)和超氧阴离子(O2.-)的含量以及过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)、超氧化物歧化酶(SOD)活性呈先升高并在不同的时间达到高峰后再下降的趋势。与箭舌豌豆相比,Cd2 胁迫下的绿豆幼苗叶组织的MDA含量、电解质泄漏率及H2O2和O2.-的积累量较高,CAT、SOD和APX的活性较低。研究表明,Cd2 胁迫下箭舌豌豆的抗氧化能力高于绿豆的抗氧化能力。

Abstract:在棉纤维加厚发育期20.0℃的日均温条件下,对棉花6~9果枝(中部果枝)和11~14果枝(上部果枝)1、2果节棉铃及其对位果枝叶分别于花后15 d和30 d,外施C/N比恒定的3种外源物质:(1)6%蔗糖和0.6%谷氨酰胺混合溶液(C+N),(2)6%蔗糖、0.6%谷氨酰胺和2μg.g-16-BA混合溶液(C+N+BA),(3)2μg.g-16-BA溶液(BA),研究低温条件下外源物质对棉株不同部位果枝棉铃纤维比强度的影响。结果表明:花后15 d外施C+N+BA可提高棉花上部果枝铃铃重和衣分,花后15 d3、0 d外施C+N+BA均可提高棉花中部果枝铃铃重和衣分。花后15 d3、0 d外施BA均有利于棉花上部果枝铃衣分的提高。花后15 d、30 d外施C+N、C+N+BA、BA均导致棉花上部果枝铃棉纤维的螺旋角和取向分布角变大,纤维比强度降低,但可优化中部果枝铃棉纤维的螺旋角和取向分布角,提高纤维比强度,且以BA处理效果最佳。

Abstract:用培养法对黄藤花粉进行了蔗糖浓度梯度、培养温度和培养时间试验;用干燥脱水法对黄藤花粉进行了低温贮藏研究.结果表明:(1)10%~15%的蔗糖浓度有利于花粉萌发生长;(2)花粉在31~35℃下培养6~9 h,其萌发生长最佳;(3)黄藤花粉随着贮藏时间的延长,花粉萌发率逐渐下降,花粉管长度较短;随着贮藏温度的加深,花粉萌发率下降趋于缓慢,花粉管长度较长。尤其在45℃下恒温干燥4 h,并在-20~-30℃下贮藏的花粉,540 d后仍然保持了较高的萌发率和较长的花粉管;(4)40℃水浴化冻和室温化冻均可用于黄藤花粉解冻,两者效果一样。

Abstract:2003~2004年对沉水樟生殖物候进行了连续2年的野外定位观测,从沉水樟的生殖繁育过程及其种子品质两方面对其有性生殖过程进行系统研究,以探讨沉水樟种群濒危机制。结果表明:(1)沉水樟的花期长达3个月,而盛花期只有15 d左右,杂交指数值为2,繁育系统类型为兼性自交,花粉胚珠比值为244.7~2 588,为风媒传粉的自花授粉植物;(2)沉水樟种子横径为0.50~0.83 cm,纵径为0.87~1.28 cm,结实量比较多,但由于不同物候期重叠,落果率高,果实产量低;(3)沉水樟的生育期长达19个月,其中果实发育期16个月,在漫长的生殖发育过程中易受环境因素影响,种子病虫危害严重,存在严重的空心现象,影响了沉水樟种群的天然更新,这是导致沉水樟种群濒危的主要原因。同时人为破坏和对现代生境不适应也是影响沉水樟种群濒危的原因。

Abstract:采用样地法对黄龙山林区实施抚育和封育措施的辽东栎林进行了调查,分析了2种培育措施对辽东栎种群径级结构、静态生命表和存活曲线的影响,并运用时间序列模型对不同培育措施的辽东栎种群数量动态进行了预测,结果表明:黄龙山林区辽东栎种群的径级结构从整体上为金字塔形,小径级个体数量较多,中、大径级个体数量较少,属于稳定型,经过抚育,辽东栎种群的径级结构更加合理;不同培育措施的辽东栎种群存活曲线都基本接近DeeveyⅢ型,种群个体早期死亡率较高,进入生理衰退年龄之后,死亡率趋于稳定;时间序列分析表明不同培育措施的辽东栎各径级株数均有不同程度的增加,径级越小增加幅度越大。根据黄龙山林区辽东栎种群的生长特点,建议在辽东栎林发育到一定时期进行抚育以创造有利的生长和发育条件,另外在种子丰产年,适时采收种子,建立苗圃,扩大人工种群。

Abstract:以研究陕西太白金矿尾矿区根瘤菌的资源多样性和重金属、苯酚抗逆性菌株的筛选为目的,选取分离自尾矿废弃地及太白河边的8种豆科植物的108株根瘤菌及42株参比菌株,进行了生长温度范围,唯一碳源、氮源利用,生长初始pH,耐盐性,重金属和苯酚的抗性等151项表型性状分析。结果表明,供试根瘤菌的表型特性存在明显差异,在85%的相似水平上供试菌株聚成5个类群,其中类群I有26株菌,主要分离自天蓝苜蓿(Medicago lu-pulina);类群Ⅱ有12株菌,其寄主各不同;类群Ⅲ有3株菌,寄主均为金翼黄芪(Astragalus chrysopterus);类群Ⅳ有40株菌,主要分离自金翼黄芪;类群Ⅴ有4株菌,寄主均为截叶铁扫帚(Lespedeza cuneata)。类群Ⅳ与已知种R.mongolense聚在一起,其这4个类群未与参比菌株聚在一起,可能是新的表观群,其分类地位需进一步确定。一些菌株表现了较强的抗重金属和苯酚毒性的能力,其中1株菌能耐受0.15 mmol/L的Hg2 ,4株菌能耐受0.4mmol/L的Cu2 ,3株菌能耐受1.6 mmol/L的Zn2 ,5株菌能耐受900 mg/L的苯酚。

Abstract:报道了中国西部赖草属一新种———贫穗赖草。该种相似于若羌赖草和广叉赖草,但它不同于前者在于穗状花序的中部或下部具孪生小穗,外稃披针形、无毛,具1.0~1.5 mm长的短尖头,内稃短于外稃1.0~1.5 mm,沿上部脊疏生小刺,叶鞘无毛,具非膜质边缘;不同于后者在于秆较高,叶片内卷,穗状花序稀疏、狭窄,小穗较短,颖较长,外稃披针形、较长,具5条不明显的脉。

Abstract:为提高甜瓜离体培养的再生率和转基因效率,以优质甜瓜品种‘伽师瓜’(‘卡拉库赛’)离体再生不定芽为外植体,通过连续多代继代培养,对引起玻璃化苗现象的几个主要因素进行了研究。结果表明,在甜瓜离体再生继代培养中,外植体继代次数是影响玻璃化发生的主要因素,同时培养基中的6-BA浓度偏高、琼脂浓度偏低以及蔗糖浓度偏低或偏高等可导致玻璃化苗的增加。培养基中较低的6-BA浓度(0~0.2 mg/L),琼脂浓度为6 g/L,蔗糖浓度为25 g/L以及添加活性炭等措施可有效地降低甜瓜玻璃化苗的发生。

Abstract:采用菌丝生长速率法研究了卷叶黄精根提取物对植物病原菌的抗菌活性。结果表明:卷叶黄精对所选10种植物病原菌中的7种呈现不同程度的抗菌活性,而且各萃取物的抑菌菌种各不相同,其中正丁醇萃取物对4种病原菌、乙酸乙酯萃取物对3种病原菌有明显的抑菌活性,水浓缩物仅对1种病原菌有抗菌活性,而石油醚萃取物对供试植物病原菌没有抗菌活性;正丁醇萃取物对苹果褐腐病、玉米大斑病、棉黄萎病病原菌抑制作用较强,EC50分别为261.26、376.03和430.05μg.mL-1;乙酸乙酯萃取物对苹果腐烂病、番茄黑霉病病原菌抑制作用较强,EC50分别为386.00和865.75μg.mL-1,各样品对植物病原菌的毒力随着浓度的增高而增强;卷叶黄精对植物病原菌的抑菌活性随着作用时间的延长,均呈现不同的下降趋势。
