Abstract:以草莓栽培品种哈尼"为材料,利用根癌农杆菌介导的叶盘法将拟南芥冷诱导转录激活因子CBF1(C-re-peat binding factor)导入草莓中.转化植株经25 mg/L Km筛选后进行PCR和Southern杂交鉴定,结果显示CBF1基因整合进草莓基因组中.采用电解质渗漏法和生长恢复法对转基因株系进行抗寒生理鉴定,结果显示,转基因株系的电导率普遍低于对照;将转基因株系和对照于-2-0℃放置7 d,在转基因株系83中有65%的植株出现了萎蔫,而对照植株有91%出现了萎蔫,经22-25℃下进行恢复生长,转基因株系83有74%完全恢复,而对照株系只有18%恢复.结果表明CBF1基因导入草莓中可以提高草莓对低温胁迫的抵抗力.

Abstract:以南丰蜜橘[Citrus reticulata Blanco var.kinokuni(Tanaka)H.H.Hu]基因组DNA为模板,根据已报道的柑橘β-胡萝卜素羟化酶基因保守序列设计4对引物,进行PCR扩增,得到4条长度为470、761、294和991 bp的片段.将这些片段克隆到pMD18-T载体,并进行测序.测序结果拼接成1条2 326 bp的序列.分析发现该序列含6个内含子,7个外显子.内含子两侧有典型的GT-AG保守序列.该序列中预测的编码序列与温州蜜柑、拟南芥等植物的β-胡萝卜素羟化酶基因序列保守性达80%以上,表明该序列确实为β-胡萝卜素羟化酶基因.该基因编码了1个含311个氨基酸的蛋白.将该序列递交到GenBank数据库,序列号为AM408552.

Abstract:以黄花补血草(Limonium aureum(L.)Hill.)无菌苗为材料,研究了不同激素配比条件下不同外植体愈伤组织的诱导及植株再生.结果表明,黄花补血草无菌苗叶片、叶柄和幼根均可作为离体培养的外植体,但叶片和叶柄的诱导率明显高于幼根.将外植体接种于分别添加0.5 mg/L NAA或1.0 mg/L 2,4-D或0.3-0.5 mg/L 6-BA 0.5-2.0 mg/L NAA的MS培养基上,经过14-28 d培养后,可脱分化产生乳白色、红色或浅绿色颗粒状或致密愈伤组织,频率达到70%以上.在MS 0.3 mg/L 6-BA 1.0 mg/L NAA培养基上,红色和绿色颗粒状愈伤组织经过1-2次继代后,均可分化产生不定芽,进而形成丛生芽,分化率达到100%.将高约3 cm的丛生芽切下,接种于分别添加0.5 mg/L IAA或0.5 mg/L IBA的1/2 MS培养基上可产生不定根,获得完整的再生植株.

Abstract:采用5种不同的水分处理,于室内生长箱内盆栽条件下对引种禾草柳枝稷(Panicum virgatum)、乡土禾草白羊草(Bothriochloa ischaemum)和栽培禾草谷子(Setaria italica)等苗期根冠生长和水分利用特征进行了比较分析.结果显示:(1)充分供水下3种禾草的苗期生物量和蒸腾效率均显著高于其它4种水分处理,而高水与低水处理下各自的根冠比无显著差异;(2)各水分处理下谷子苗期总生物量、蒸腾效率和耗水量均显著大于白羊草和柳枝稷;(3)在5种水分处理下,苗期根冠比谷子均最小,柳枝稷最大;(4)低降复水后,3种禾草生物量和蒸腾效率较低水处理分别显著提高16.7%-98.7%和28.2%-118.2%,显示出补偿效应,以白羊草增幅最大.结果表明,白羊草、谷子和柳枝稷在不同土壤水分处理下的苗期生物量、根冠分配比例以及水分利用效率差异反映了野生种、栽培种和引进种禾草苗期对半干旱环境条件水分生态适应性的异同.

Abstract:对麦秸翻压还田后隔茬冬小麦根系活力、旗叶叶绿素含量、丙二醛(MDA)含量及光合速率等衰老指标进行了测定分析.结果显示,(1)小麦秸秆还田后可显著促进各时期根系的生长并显著提高小麦根系活力,且根系在30cm处土壤中的分布增加;(2)冬小麦生育后期叶片叶绿素以处理Ⅲ(6 000 kg/hm2)的降解速度(18%)最慢,旗叶光合速率在灌浆期和蜡熟期以处理Ⅲ为最高,旗叶丙二醛含量以处理Ⅲ最低,比对照(Ⅰ)低6.78 mmol·g-1FW,丙二醛含量与光合速率呈显著负相关.研究结果表明,适量秸秆还田对接茬玉米的产量影响不显著,但可显著延缓隔茬冬小麦的衰老并显著提高产量,在黄土高原有灌溉条件的地区以6 000 kg/hm2的小麦秸秆量还田较适宜.

Abstract:从菊科植物大花金挖耳(Carpesium macrocephalum)的花、茎、叶、果实和根中分离出92株内生菌,其中真菌68株,细菌16株,放线菌8株;对峙法生物活性测定结果表明,其中22株内生菌分别对小麦赤霉病菌、黄瓜灰霉病菌、辣椒疫霉病菌、玉米大斑病菌和苹果炭疽病菌均有较好的拮抗作用;采用生长速率法测定22株内生菌发酵产物的抑菌活性,结果表明,内生细菌H-32的发酵液稀释10倍和内生真菌H-18、Y-12、G-4、G-6的菌丝丙酮提取物在0.5 mg·mL^-1(菌丝干重)的剂量下,均对番茄灰霉病菌和玉米大斑病菌菌丝生长具75%以上的抑制效果;活体组织法测定结果,H-32的发酵液和H-18、J-1、Y-17、G-6的菌丝丙酮提取物在2.0 mg·mL^-1(菌丝干重)的剂量下,对番茄灰霉病的防治效果均在50%以上.综合分析认为,H-32、H-18和G-6等内生菌的抑菌作用值得进一步研究.
