Abstract:应用光学显微镜、紫外分光光度计和傅立叶显微红外光谱技术,对不同CO2浓度(370、700μmol?mol-1)处理条件下,对拟南芥茎秆的形态结构及化学成分进行解剖学和组织化学的对比研究.结果发现,在CO2浓度升高后,茎秆的直径、面积、形成层细胞活性及维管束面积等均有所增加;化学成分分析显示,茎秆中纤维素、木质素以及可溶性糖的含量均比对照有所提高,且纤维素和可溶性糖增加显著.研究表明,拟南芥茎秆的解剖结构及化学成分对大气CO2浓度升高表现出正响应,从而为提高茎秆水分和营养物质的运输以及对生殖器官的支撑等各项功能提供更多的结构和物质基础.

Abstract:以优化的毛白杨无性系85号最适遗传转化体系为基础,通过农杆菌介导法将Bt(CryⅠAc)和API双价抗虫基因导入毛白杨无性系85号基因组.在高浓度卡那霉素的培养基上诱导不定芽和根,初步选择到200株转化植株,PCR检测显示,66株呈阳性反应.Southern杂交和ELISA检测进一步证明,Bt(CryⅠAc)和API双价抗虫基因已整合到毛白杨无性系85号基因组中,并得到了表达.对转化植株用舞毒蛾幼虫进行饲虫试验,结果显示昆虫幼虫的死亡率高达60.0%~77.8%,且存活幼虫的生长发育也受到了明显抑制.研究结果为杨树抗虫育种提供了新的种质资源.

Abstract:芪合成酶是白藜芦醇生物合成过程中起关键作用的酶.通过RT-PCR,从高抗葡萄黑痘病的中国野葡萄毛葡萄商-24克隆了芪合成酶基因家族的2个成员VqSTS1和VqSTS2,其cDNA全长均为1 179 bp,编码392个氨基酸.氨基酸序列分析表明,VqSTS1和VqSTS2编码氨基酸247-257位点的IPN(S/F)AGAIAGN "是芪合成酶家族固有的氨基酸保守结构域,368-378位点的GVLFGFGPGLT"是芪合成酶与查尔酮合成酶家族固有的保守序列特征.在编码的392个氨基酸中,VqSTS1和VqSTS2仅有12个氨基酸不同,氨基酸一致性达97%.多重比较分析显示,VqSTS1与VqSTS2与五针松、河岸葡萄、白粉藤、华东葡萄及欧洲葡萄等STS在氨基酸水平的一致性分别为65%、96%、97%和98%.将VqSTS1和VqSTS2登录GenBank,登录号分别为EF158856和EF158857.

Abstract:以二倍体、四倍体和六倍体冰草(P基因组)以及中国春、Fukuho、栽培一粒小麦和硬粒小麦(ABD基因组)等为材料进行RAPD分析,从520个RAPD随机引物中,筛选出2个P基因组特异的RAPD分子标记OPC04和OPP12.利用OPC04和OPP12对小麦族其它基因组植物和8个小麦-冰草二体附加系进行扩增,结果表明OPC04和OPP12在ABD、C、E、AG、I、M、R、S、V、Y等基因组扩增中未出现相应结合位点;而在对8个小麦-冰草二体附加系扩增中均出现此2个特异片段,且扩增稳定、重复性好.进一步表明OPC04和OPP12是冰草P基因组的特异标记,可作为小麦-冰草重组系外源P染色质的鉴定标记之一.

Abstract:三萜皂甙是中药三七(Panax notoginseng)的主要有效成分,为阐明三七三萜皂甙合成途径中法呢基焦磷酸合酶(FPS)的结构特征,采用RT-PCR及3-'RACE和5′-末端分析法,对三七FPS进行克隆,获得三七FPS cDNA全长,已提交GenBank,登录号为DQ059550;结合生物信息数据库及相关分析软件,对三七根FPS进行性质预测,并与GenBank中记载的植物FPS进行比对分析,构建了植物FPS系统进化树,进行聚类关系分析,根据FPS聚类关系将这些植物异戊二烯类物质合成途径的产物分组.结果表明,植物FPS有2个保守核心区,分别为DDIMD和QVQDDYLD;比对分析及构建的系统进化树表明,三七与人参、积雪草的FPS序列一致性分别为99%、95%,其三萜物质结构也相似.

Abstract:用染色体步移技术(chromosome walking)的基本原理以辐射松(Pinus radiata)肌动蛋白基因(actin)为例,利用获得的EST序列设计定向引物,向上游和下游进行了染色体序列的步移.获得了包括启动子、5′端非编码区和编码区及3′端非编码区辐射松肌动蛋白基因基因组序列2154 bp.通过对200株不同辐射松个体进行PCR扩增及测序,共获得了21个SNPs,其中启动子区域3个,编码区15个,3′端非编码区4个.实验结果为今后染色体步移技术在基因非编码区SNP的检测提供了理论与技术参考.

Abstract:针对灰木相思(Acacia implexa Benth.)蛋白质提取过程中含有大量色素和酚等干扰物质的现象,进行了实验条件的优化,结果显示:选择含较少次生物质量的组培苗为实验材料更易得到清晰的双向电泳图谱,在裂解液中添加硫脲更有利于提高蛋白溶解度,第一向等电聚焦固相pH梯度胶条(24 cm)的最适蛋白上样量为1000μg,表明采用优化后的双向电泳体系提高了灰木相思总蛋白双向电泳的分辨率和重复性,建立起一套适用于灰木相思总蛋白质组分析的双向电泳(2-DE)方法.

Abstract:在田间条件下,选用3个小麦材料与不同玉米品种进行杂交,用含不同体积分数二甲基亚砜(DMSO)的100 mg/L 2,4-D溶液和3%DMSO与2,4-D配比处理两种方法诱导小麦×玉米产生单倍体.结果表明,在不同DMSO处理中,2%体积分数的DMSO最佳处理平均得胚率最高为15.8%,3%处理最低(7.1%),二者差异显著;在激素配比试验中,以授粉后48 h采用小花滴注100 mg/L 2,4-D溶液、授粉后72 h再次采用小花滴注并穗下茎节注射3%DMSO的100 mg/L 2,4-D溶液的处理得胚率最高(14.1%),但与3%DMSO的2,4-D 1次处理(13.4%)2、,4-D 1次处理(12.7%)、2,4-D 2次处理(10.6%)和3%DMSO的2,4-D 2次处理(12.6%)之间无显著差异,而这5种处理的得胚率均与对照和未授粉只进行3%DMSO的2,4-D 1次处理存在极显著差异.

Abstract:在大田试验条件下,研究了不同施氮水平对2种穗型冬小麦品种花后干物质和氮素积累与运转的影响及其与产量和品质的关系,以探讨氮素营养水平对冬小麦碳氮运转的影响.结果显示,适宜的施氮量(180 kg·hm^-2)能够极显著增加2种穗型冬小麦品种叶片、茎鞘等营养器官花前贮藏物质及花前贮藏氮素的再运转量和运转率以及总再运转量和运转率,也能够极显著增加成熟期籽粒氮素含量和花前贮藏氮素总运转量对籽粒氮素含量的贡献率.各施氮处理对2种穗型小麦品种花后氮素积累量对籽粒氮素含量贡献率的影响效应不明显.结果表明,适宜的施氮量有利于小麦籽粒和蛋白质产量的提高.

Abstract:采用常压硅胶柱层析和RP-HPLC制备并结合杀虫、抑菌活性追踪的方法,从博落回中分离获得了杀灭金鱼指环虫和抑制鱼类病原菌的活性成分,通过TLC、UV、m.p.、IR、MS、1H NMR和13C NMR鉴定其为血根碱.杀灭指环虫试验结果表明:血根碱对指环虫有较强杀灭活性,在质量浓度为0.6 mg?L-1时的杀虫率达到100%.血根碱对6株水产致病菌的活性试验结果表明:对嗜水气单胞菌的活性最高,MIC和MBC分别为12.5和25μg·mL-1;对哈维弧菌、鳗弧菌、杀鲑气单胞菌史氏亚种的活性最弱,其MIC和MBC均分别为50和100μg·mL-1.血根碱杀灭鱼类指环虫活性较高,对6株水产病原菌都有明显抑制作用.

Abstract:以具4种细胞质、2种核类型的粘类小麦雄性不育系为测验种,对309份来自国内外不同地区普通小麦品种对应粘类小麦不育系的育性恢复和保持关系进行调查,研究粘类小麦雄性不育系育性基因的地理分布.结果表明:(1)普通小麦品种中广泛存在着粘类雄性不育系的恢复基因,所配组合中,高恢复度以上的组合占到45.24%;(2)所配组合F1小穗结实率范围在0~91.35%之间,其中0和60%~80%的分布范围较多;(3)具有恢复能力的品种在供试6个地区均有分布,但比例不同,中国南方、新疆内陆、青藏高原等春麦地区高,可育品种的分布比例均超过50%;具有育性保持作用的品种在中国黄淮海暖温带冬麦气候生态区和加拿大地区分布较多,在新疆麦区分布最少;(4)恢复度80%以上的品种在6个地区均有分布,但在各地区供试材料中的比例都不高.

Abstract:对矮沙冬青(Ammopiptanthus nanus Cheng f.)的形态学特征、开花物候及花空间分布进行了研究,结果表明:在单花、花序、单株、居群水平上的开花物候都表现出很强的开花不同步性,在单花、单花序水平主要表现在单花寿命4~9 d不等,平均(6.74±1.23)d,以7 d出现的频率最高;单花序开花持续时间范围在4~13 d,平均(8.33±2.63)d,以10 d出现的频率最高;单株及居群水平上表现更为明显.矮沙冬青的花具有虫媒传粉的特征,花主要分布在花枝的上部,下部花所占比例很小,有很多花枝花全部为上部花.相关性分析表明上部花和总花数相关性极显著(r=0.990,P<0.01),而中部花、下部花和总花数的相关性不显著,甚至呈负相关.单株不同方位开花数量差异不显著(F=0.0579,P>0.05).
