Abstract:通过RACE技术,克隆了云南红豆杉紫杉烷2α-O-苯甲酰转运酶基因(TyTBT),该酶催化2-去苯甲酰-7,13-二乙酰巴卡亭Ⅲ生成7,13-二乙酰巴卡亭Ⅲ,是紫杉醇合成途径中的关键酶之一.TyTBT基因cDNA全长1481 bp,含有1 320 bp的开放读码框,编码440个氨基酸的多肽,分子量为50 050 Da,等电点为6.17.氨基酸序列比对表明TyTBT同植物酰化酶家族的其它成员有较高的相似性,超过67%,同东北红豆杉和曼地亚红豆杉的紫杉烷2α-O-苯甲酰转运酶氨基酸序列的一致性和相似性达到最高,分别为95%和96%.广泛地比对分析证明这种较高的相似性在红豆杉属的其它酶家族中具有普遍性,进化树分析表明同东北红豆杉和曼地亚红豆杉的紫杉烷2α-O-苯甲酰转运酶(TBT)的相似性高于紫杉醇合成途径中的其它酰化酶.

Abstract:采用花粉介导方法将几丁质酶基因和潮霉素基因导入玉米(Zea maysL.)自交系海92-1中,以筛选抗玉米丝黑穗病的转基因品种.对转化植株及其后代植株的PCR、Southern blot检测表明,目的基因已导入转化植株并整合到其基因组中,且能够稳定遗传.ELISA分析证明转基因植株中目的基因可高效表达,表达产物量在9.8~16.3 ng?g-1鲜叶左右.统计分析显示,目的基因产物表达水平与转基因植株的抗病性呈极显著正相关(r=0.925,P<0.01).接种病毒鉴定结果揭示转基因株系的抗病性比对照提高3~4级.结合农艺性状筛选,选育到401、403这2个抗丝黑穗病且其它农艺性状优良的转基因纯合株系.

Abstract:采用RNAi抑制NHX1基因家族的表达,并观察其对拟南芥耐盐性和耐旱性的影响.根据从拟南芥(Ara-bidopsis thaliana)cDNA中扩增出编码Na /H 反向转运蛋白基因AtNHX1长度为210 bp高度保守序列作为RNAi的靶标区,并正反2个方向插入载体pHANNIBAL中,2个片段用intron连接;将RNAi表达框连入具有NPTⅡ筛选标记基因的表达载体pART27中,构建以拟南芥NHX1基因家族为靶标的RNAi载体.采用农杆菌介导的真空渗透法转化拟南芥,得到T0代转基因拟南芥种子.对转基因阳性植株进行RT-PCR检测以及耐盐性和耐旱性分析.结果表明,利用本实验构建的NHX1基因家族RNAi载体,拟南芥NHX1基因家族表达被成功地抑制;耐盐和耐旱分析表明RNAi技术对基因表达沉默是有效的.

Abstract:采用SD-PAGE方法对二色胡枝子14个居群147个个体单粒种子盐溶蛋白进行了电泳检测,共检测到28个蛋白单体,其中多态性单体23个,多态百分率为82.14%,各居群内多态百分率平均为33%。各居群间的遗传分化系数(Gst)为0.4669。居群间基因流(Nm)为0.5709。种群内的基因多样性占总群体的53.3%,表明二色胡枝子种群具有丰富的遗传多样性,居群内变异是二色胡枝子遗传多样性的主要来源。聚类分析结果将14个二色胡枝子居群分为3类,居群间地理距离和遗传距离相关性不显著。

Abstract:以辣椒细胞质雄性不育系21A及其同核异质保持系21B为试验材料,比较分析两系雄配子发育过程中酯酶(EST)、谷氨酸脱氢酶(GDH)和苹果酸脱氢酶(MDH)同工酶的表达特征。结果表明:幼叶和花蕾的EST同工酶酶谱在谱带数目和酶带强弱上存在时空表达差异,并且随着雄配子发育的进行,保持系21B从中花蕾至特大花蕾比不育系21A多1条清晰的谱带(EST3e),其差异表达发生在细胞学上观察到的败育时期之前;在GDH同工酶中,保持系21B从大花蕾至特大花蕾比不育系多6条谱带(GDH,和GDH1/2),酶谱差异表达时期与细胞学上观察到的败育时期一致;而在MDH同工酶中,不育系21A和保持系21B的幼叶和各级花蕾的酶谱在谱带数目和谱带强弱上均没有明显差异。

Abstract:利用荧光显微镜对百合远缘杂交组合Bernini×Pollyanna花粉萌发及花粉管生长过程进行观察研究,结果显示,授粉后3~30 h内花粉萌发形成花粉管,且花粉管生长速度由快到慢,48~51 h内花粉管停止生长,花粉管最后深入到花柱的1/3处,并观察到一些异常的花粉管形态;花粉管生长过程中还伴随着一系列的胼胝质反应,出现的部位依次是乳突细胞、花粉管、花柱通道细胞、胚珠中的胚囊.结果表明该杂交组合不亲和.研究认为Berni-ni柱头乳突细胞是阻碍Pollyanna花粉管生长的第一道屏障,花柱通道细胞是抑制花粉管在花柱内生长的第二道屏障.

Abstract:在大田栽培条件下,以2个不同穗型的高产冬小麦品种为试验材料,在施240 kg·hm-2(N240)和330 kg·hm-2(N330)纯氮水平下,分别施纯硫60 kg·hm-2(S60)和0 kg·hm-2(S0),研究了施硫对小麦不同器官碳氮运转及其对籽粒产量和蛋白质产量的影响.结果显示:(1)2个供氮水平下,施硫(S60)均比对照(S0)增加了2个小麦品种的叶片、茎、鞘、颖壳和穗轴等营养器官花前贮藏干物质、氮素的运转量和运转率以及总运转量和总运转率,提高了转运干物质、氮素对籽粒重和籽粒氮素的贡献率,极显著提高了籽粒和蛋白质产量.(2)施硫对豫农949品种籽粒和蛋白质产量提高幅度显著高于兰考矮早八;与对照(S0)相比,豫农949的N240S60和N330S60处理使籽粒产量分别增加12.74%和16.41%,蛋白质产量分别增加16.84%和16.14%.结果表明,中氮和高氮水平下施硫均可明显促进高产小麦植株的C-N运转,提高植株对氮的吸收利用和碳物质的积累,从而增加籽粒产量,但不同品种间的施硫效应存在差异.

Abstract:通过盆栽控水试验,分析了8种美国引进禾本科牧草在不同水分胁迫下叶片的叶绿素、脯氨酸、可溶性糖含量和叶绿素荧光参数等的变化.结果表明,叶片叶绿素相对含量总体呈随水分胁迫程度加剧而降低的趋势,并在严重胁迫(HWS)下达到最低;在轻微水分胁迫(SWS)下各牧草脯氨酸含量增加缓慢,随水分胁迫的进一步加剧而急骤升高;可溶性糖含量随水分胁迫的加剧而持续增加;叶绿素荧光参数Fv/Fm、Fv/Fo、ΦPSⅡ均随水分胁迫加剧先升高后降低,在轻度水分胁迫下达到最大,但各草种间变异情况不同.运用隶属函数法所得牧草综合抗旱性强弱顺序为:Ⅳ>Ⅰ、Ⅷ>Ⅱ>Ⅴ>Ⅲ>Ⅵ>Ⅶ,运用萎蔫系数直接评价的抗旱性结果为:Ⅳ>Ⅷ>Ⅰ>Ⅱ>Ⅲ>Ⅴ>Ⅵ>Ⅶ,即猫尾草(Ⅰ)、无芒雀麦(Ⅳ)和新麦草(Ⅷ)抗旱性较强,而细茎披碱草(Ⅵ)、高冰草(Ⅶ)较差.

Abstract:以NC89烟草品种为材料,用白纱布遮荫研究了100%、80%、50%和20%全光照处理对烤烟生长发育、光合特性及化学成分的影响.结果表明:(1)随光照强度的减弱,植株茎杆变细,叶片的长宽比增加,叶片数减少,出叶速度变慢,干物质重逐渐减少,并以80%的全光照条件下的株高、叶面积最大.(2)净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)、叶绿素a/b值均随光照减弱而逐渐下降,胞间CO2浓度(Ci)、叶绿素含量逐渐升高;叶绿素荧光参数Fm、ФPSⅡ、Fv/Fm、QP值逐渐增加.(3)烟碱、淀粉、还原糖、水溶性总糖、总酚含量随光照减弱而降低,而总氮和钾含量升高.综合分析各项测定指标发现,在大于或等于80%全光照(光照强度为770~1 125μmol?m-2?s-1)条件下,烤烟的生长指标变化不大,品质指标符合不同卷烟工业需要,适合优质烟叶的生产.

Abstract:从抗寒性、光合特性、生长及其结果性状等方面研究了宁夏贺兰山东麓酿酒葡萄的生态适应性。室内抗寒性鉴定和田间抗寒评价结果表明:‘歌海娜’最不耐寒,抗寒性强的品种有‘长相思’、‘雷司令’、‘黑比诺’、‘佳美,;抗寒力较差的品种有‘赛美容’、‘白诗南’、‘白玉霓’、‘神索’;抗寒力较强的品种是:‘霞多丽’、‘赤霞珠,、‘梅鹿辄’、‘白比诺’、‘西拉’。对酿酒葡萄的光合特性的测定发现:同一立地不同品种的净光合速率(Pn)差异明显,‘赤霞珠’的光合速率远高于‘霞多丽’、‘梅鹿辄’的,各品种光合速率的日变化呈双“S”曲线;全天光合速率的最高峰出现在上午10:00左右,下午14:00出现光合低谷,次高峰出现在下午17:00左右。同一品种在不同立地上的叶面积(LA)、叶鲜重(LFW)、比叶重(SLW)、叶绿素(Ch1)含量均表现差异;不同品种在同一立地条件下以上几个指标也表现不同。酿酒葡萄在贺兰山东麓不同区域的生长结果表现也不同。研究表明:大多数酿酒葡萄品种在贺兰山东麓具有良好的生态适应性,土壤和品种是造成贺兰山东麓酿酒葡萄生态适应性差异的主要因素,该区酿酒葡萄栽培的首要任务是解决抗寒栽培技术和土壤培肥。

Abstract:用Li-6400便携式光合测定系统对5个虎杖材料的光合生理特性日变化及其与气象因子关系进行了研究。结果表明:(1)虎杖的净光合速率日变化呈‘单峰’型曲线,日最大净光合速率(15.0μmol·m^-2·s^-1)值出现在9:00左右;(2)叶片水压亏缺、气孔导度和蒸腾速率的峰值在同一时间出现(13:00),胞间CO2浓度不随气孔导度的降低而减小,控制虎杖光合速率因子为非气孔限制;(3)供试5个材料的净光合速率日变化趋势基本一致,并以地栽组培苗的净光合速率最高,而‘贵州凯里’的最低。光合有效辐射对光合特征参数的变化影响最大,且对净光合速率起决定性作用(r-0.534^**),环境因子主要通过对蒸腾速率、叶片水压亏缺和叶面温度的作用来影响虎杖叶片净光合速率。
