Abstract:以‘条中32’接种前后的小麦抗条锈病种质‘陕麦139’幼苗叶片为材料,利用抑制消减杂交技术,构建了条锈菌接种48 h小麦抗病品种叶片的抑制表达SSH-cDNA文库,通过测序以了解条锈菌侵染后被抑制表达的相关基因。从构建的文库中随机选取48个阳性克隆,进行测序,经过序列拼接等,获得高质量EST序列34条(Gen-Bank序列号为EL930132-EL930165),序列比对分析表明其中31条EST序列与已知功能的基因序列同源性较高,主要涉及植物的能量代谢途径、膜转运、信号传导及核酸加工等方面的基因,包括光合系统Ⅱ抗性蛋白D1、碳酸酵素、线粒体磷酸盐转运子、苹果酸脱氢酶、过敏性诱导反应蛋白和rRNA核酸内切酶等相关基因。此外,获得未知功能的EST序列3条。研究表明,小麦在条锈病菌侵染初期,许多基因的正常表达调控受到影响,如能量代谢和寄主细胞结构木质化,阻止小麦抗病过敏性反应等。

Abstract:以非单性结实的全雌黄瓜为实验材料,利用cDNA-AFLP技术比较了授粉前后黄瓜幼果的基因表达差异。以ASE-AT/TAQ-CAG为引物对从授粉后黄瓜幼果组织中分离到1条特异片段,该片段仅在授粉后黄瓜幼果组织中表达,将该片段回收测序并翻译成氨基酸序列,用blastp程序在NCBI GenBank数据库中进行同源性检索和相似性比对,结果发现该片段推导的氨基酸序列与黄瓜扩张蛋白CsEXP1~9的相似性依次分别为71%、58%、63%、75%、85%、82%、67%、68%和85%,可能是一个新的黄瓜扩张蛋白基因,命名为CsEXP10,表明扩张蛋白基因可能与黄瓜果实膨大生长有密切关系。

Abstract:从除虫菊(Pyrethrumcinerariifolium Trev.)的根、茎、叶和花中分离出128株内生真菌。生长速率法测定结果表明,有56株内生真菌发酵液10倍稀释液至少对供试的番茄灰霉病菌(Botrytis cinerea)、辣椒疫霉病菌(Phytophthora capsici)、苹果炭疽病菌(Colletotrichum gloeosporioides)、玉米大斑病菌(Exserohilumturcicum)、小麦纹枯病菌(Rhizoctonia cerealis)和小麦赤霉病菌(Fusarium graminearum)等6种病原菌中的一种抑菌活性达75%以上;活体组织法测定结果,Y1、Y2、H2共3株菌的发酵液对番茄灰霉病的防效达到50%以上;盆栽试验结果表明,Y1、Y2、Y7、H2、G13共5株菌株发酵液对辣椒疫霉病的防治效果达到50%以上。综合分析认为,Y1、Y2、H2、G13等内生真菌的抑菌作用值得进一步研究。

Abstract:以不同浓度的大蒜秸秆水浸液对西瓜枯萎病菌、黄瓜枯萎病菌、辣椒疫霉病菌进行抑菌圈试验,并对3种不同的大蒜秸秆萃取液中的可挥发抑菌物质进行鉴定,以了解大蒜秸秆水浸液的抑菌作用。结果表明,大蒜秸秆水浸液对3种靶标菌均有抑菌活性,其最有效的抑菌浓度为100 g.L-1,对辣椒疫霉病菌的抑菌活性最强。大蒜秸秆水浸液的石油醚、三氯甲烷和乙酸乙酯3种有机溶剂萃取相对3种靶标菌均有抑菌活性,以乙酸乙酯相抑菌活性最强,在50 g.L-1浓度下对辣椒疫霉菌抑菌率达100%,在100 g.L-1浓度下对西瓜枯萎病菌和黄瓜枯萎病菌的抑菌率也达100%;三氯甲烷相的抑菌活性稍高于石油醚相;抑菌活性基本随浓度增加而增大。经GC-MS分析,初步确定大蒜秸秆水浸液中的可挥发抑菌物质主要有2-甲氧基-1,6-己二酸甲酯、邻苯二甲酸二丁酯等有机酯类,3-甲氧基-4-羟基-苯甲酸、对羟基苯甲酸等有机酸,以及对甲氧基苯酚等酚类化合物。

Abstract:通过水培试验,研究了模拟酸雨(pH 3.5)与Al(3、30 mg/L)、Pb(25、250 mg/L)、Cd(0.11、mg/L)单独或复合胁迫对小麦幼苗生长及抗氧化酶活性的影响。结果表明,模拟酸雨与Al、Pb、Cd单独或复合处理12 d后,小麦幼苗根长降低54.01%~94.81%,苗高降低18.88%~77.95%,单粒萌根数降低21.87%~80.37%,其生长发育受到显著抑制,抑制效应随处理浓度增大和作用时间延长而增强,且复合处理组抑制效应更强;幼苗根部超氧化物歧化酶(SOD)活性在模拟酸雨与单一金属离子处理下显著升高,而在3种金属离子复合处理下显著降低,但酸性条件下无论金属离子单独及复合处理后,过氧化物酶(POD)活性均显著升高,过氧化氢酶(CAT)活性均明显下降。研究发现,酸性条件下3种金属离子对植物细胞均产生氧化胁迫作用,且它们复合胁迫的毒性效应大于其单独胁迫。

Abstract:以‘大西洋’、‘夏波蒂’2个品种的脱毒微型种薯为试验材料,分别在冷藏(4~6℃)、窖藏(8~12℃)和常温(18~22℃)条件下贮藏110 d,研究不同贮藏条件和温度对马铃薯微型种薯活力及生理特性的影响。结果表明:(1)低温冷藏能显著提高微型种薯的活力,2个品种的活力指数分别比窖藏和常温贮藏高20.44%、20.28%和33.51%、35.46%。(2)冷藏与窖藏和常温贮藏相比,CAT活性高35.37%~48.25%、91.68%~125.23%,SOD活性高5.79%~6.60%、10.53%~12.81%,而MDA含量却降低30.47%~47.79%、85.54%~87.09%。(3)随着微型种薯的重量增加其活力指数显著提高,CAT和SOD活性也增加,而MDA含量逐渐降低。(4)经不同条件贮藏后,微型种薯的含水量呈下降趋势,以冷藏条件的含水量降低最少;可溶性蛋白含量显著增加,冷藏比窖藏和常温贮藏分别高14.63%~24.67%和25.5%~35.04%;淀粉含量表现为不同程度降低,以窖藏降低最少;可溶性糖含量虽有明显降低,但其含量变化无显著差异。说明低温贮藏可以使马铃薯微型种薯保持较高的活力,适合贮藏条件的顺序依次为:冷藏>窖藏>常温贮藏。

Abstract:采用Li-6400光合分析仪,在自然条件下对不同坡位胡枝子叶片净光合速率(Pn)、蒸腾速率(Tr)及其相关环境因子的日进程进行了观测。结果表明:在5月份晴天条件下,不同坡位胡枝子叶片Pn、Tr日变化均呈单峰曲线,而水分利用效率(WUE)表现出早晚高、中午低的变化趋势。它们的叶片Pn峰值不同坡位均出现在11:00,而且上坡位峰值和日平均值略高于下坡位和平地(P>0.05);Tr峰值上坡位出现在11:00,下坡位和平地均出现在13:00,且上坡位峰值和日平均值显著低于下坡位和平地(P<0.01);上坡和平地WUE均在15:00达到最大值,但下坡位推迟到17:00,而且上坡位峰值和日平均值略高于下坡和平地(P>0.05)。胡枝子叶片Pn与PAR、Ca、RH等生态因子间的拟合方程分别为:Y上pn=-26.148 0.002 3xPAR 0.11xCa-0.84xRH,Y平pn=-19.148 0.001 9xPAR 0.106xCa 0.86xRH,Y下pn=-27.148 0.002xPAR-1.16xRH,其主要生态影响因子及其大小顺序为XPAR>XRH>XCa。研究发现,不同坡位胡枝子叶片主要光合指标日变化趋势相似,但其峰值出现时间和高低有别,其主要生态影响因子为光合有效辐射、大气相对湿度和大气二氧化碳浓度。

Abstract:对青海省民和、西宁和湟源3个地区的药用植物薄荷(Mentha haplocalyx Briq.)的净光合速率和叶绿素荧光参数的变化进行分析比较。结果显示:(1)最大净光合速率(Pn)湟源>西宁>民和;(2)PSⅡ反应中心最大光化学效率(Fv/Fm)以及PSⅡ反应中心活性参数(1/Fo-1/Fm)和实际光化学量子效率(ΦPSⅡ)的变化趋势相同,均以西宁最高,而相应的光合功能的相对限制(LPDF)西宁最低;(3)西宁薄荷的光化学猝灭系数(qP)最高(0.875),而非光化学猝灭(NPQ)则最低(1.114)。研究表明,青海薄荷有光抑制现象发生,但并没有造成PSⅡ反应中心的不可逆破坏;西宁薄荷光化学能力较高而湟源薄荷对于过剩光能的耗散能力较高。

Abstract:通过对Amana anhuiensis主模式的存放地中国科学院植物研究所标本馆(PE)的查找及向原作者证实,确认该种的模式已不存在。在对模式产地的野外调查过程中,发现仅有这一种老鸦瓣分布,且其花期及果实成熟期植株的一些重要形态性状与原描述不符或在原描述中没有提及,这些性状不仅在个体间比较稳定,而且在种的划分上具有重要价值。它们是:鳞茎皮黄褐色,薄纸质,内侧被绵毛;第1叶自基部向上3/4处最宽;苞片狭披针形;花被片内侧基部具黄绿色斑块;内轮3枚雄蕊比外轮3枚长,花粉黄色,雌蕊花柱与子房近等长。朔果近球形,果喙长10 mm左右。为此,我们指定了该种的新模式,并对其形态特征进行了补充描述。
