Abstract:通过对新选育的甘蓝型油菜高温敏感生态型不育系417S与30份中外品种(系)进行测交、杂交和回交,研究其温敏不育性的遗传特点.结果表明:(1)30份品种(系)可不同程度的恢复417S的育性,其中24个品种(系)可完全恢复417S育性,占所配组合的80.0%;5个品种(系)可高度恢复417S的不育性,占所配组合的17.2%.(2)自然条件下同一组合正反交F2群体育性分离结果不同,人工控温条件下不同组合F2分离群体中可育与不育呈现15∶1分离,回交群体中出现3∶1分离,表明417S温敏不育性受细胞质和2对隐性重叠核基因共同控制.(3)以417S与1521C构成的回交一代群体为试材,采用分离群体分组分析法(BSA法)对回交一代群体构成的不育集团和可育集团筛选育性基因的RAPD标记,从247个供试引物中筛选出2个引物BA392(5′-AGTCACTCCC-3′)、S113(5′-GACGCCACAC-3′)在不育群体和可育群体间扩增出多态性产物.经进一步对F2分离群体的单株检测,引物BA392扩增出了得到重复验证和可靠的多态性扩增产物,其产生的特异条带BA392-400bp与417S温敏雄性不育的恢复基因连锁,遗传图距为6.0cM,可作为417S温敏不育恢复基因的连锁标记.

Abstract:以金黄色非洲菊品种深圳5号(S5)为材料,探讨了利用羟脯氨酸(Hyp)筛选耐热变异系的方法,初步建立了非洲菊耐Hyp变异系的离体筛选体系.结果表明,非洲菊新鲜花托和芽适宜的Hyp初始筛选剂量为0.4mg·mL-1,而预培养25d花托的初始筛选浓度宜增加到0.6mg·mL-1;获得的存活芽恢复培养后进行后续筛选时可以选择0.5~0.6mg·mL-1Hyp;通过对花托和芽进行5次间隔筛选后获得耐Hyp变异系SHP5,得率为0.7%.与对照(S5)相比,SHP5在室内高温胁迫条件下具有较高的游离脯氨酸含量,在自然高温条件下长势好,花序梗长度和花色素苷含量均显著增加,表现出较强的耐热性.

Abstract:以黄河蜜幼苗为材料,分析了化学诱导物处理后甜瓜叶片中几丁质酶(CHI)、β-1,3葡聚糖酶(GLU)、苯丙氨酸解氨酶(PAL)和多酚氧化酶(PPO)活性的变化.结果显示:经5种化学诱导物处理后,甜瓜叶片PAL活性快速增加,于处理后第6天出现活性高峰;苯丙噻二唑(BTH)、水杨酸(SA)、草酸(OAA)和硅酸钠(Na2SiO3)处理后GLU活性变化与PAL相似,PPO活性在测定期内持续升高;磷酸氢二钾(K2HPO4)处理对GLU和PPO活性无明显影响;SA处理后叶片CHI活性明显升高,其它诱导物处理对CHI活性无明显影响.结果表明,防卫酶(特别是GLU和PPO)活性表达增强是BTH、OAA、SA和Na2SiO3等诱导物处理系统诱导甜瓜幼苗白粉病抗性增强的重要生化机制.

Abstract:以离体香樟叶片和果实为材料,研究了萘乙酸(NAA)处理对其中酪氨酸酶及其同工酶活性的影响,并对香樟果实中酪氨酸酶进行分离纯化,初步分析了该酶基本生化特征.结果表明:(1)10μmol·L-1NAA处理对香樟叶片同工酶TYRL2有较强的激活作用,对同工酶TYRL1有抑制作用,1.0和0.1μmol·L-1浓度的NAA对同工酶TYRL3有激活作用;(2)在香樟果实的酪氨酸酶同工酶中,TYRF1的活性最高并在果实成熟过程中逐渐增强,100μmol·L-1的NAA对果实成熟以及酪氨酸酶的酶活性都有一定的促进作用;(3)经纯化的香樟果实酪氨酸酶最适pH值为7.5,最适温度为50℃,亚基分子量为43KDa;以L-DOPA为底物时的Km为12.82mmol·L-1,Vmax为80.65U·mg-1蛋白;其在酸性(pH4~7)和高温(40~70℃)条件下较稳定.可见,不同浓度的NAA对香樟叶片和果实中的酪氨酸酶活性有不同程度的激活和抑制作用.

Abstract:以半蔓生型奶花芸豆(Y09)和直立型奶花芸豆(Y06)为材料,通过SDS-PAGE分析研究了开花后种子形成过程中贮藏蛋白(SP)的积累规律.结果表明:2个芸豆品种开花后种子形成过程中,子叶、胚蛋白积累变化趋势基本一致,但其变化幅度存在显著差异,其中,在开花后20d内,半蔓生型芸豆Y09的籽粒蛋白积累量大于直立型芸豆Y06,但在开花后25~40d,Y06籽粒蛋白积累量显著高于Y09;两芸豆品种的子叶和胚均含有丰富的贮藏蛋白(SP)(14.4~97.4kD),其中子叶的41.0、43.9、39.4kD3种蛋白含量较高,约占子叶总蛋白含量(光密度)50%以上,而胚蛋白亚基分布相对均匀;两品种的子叶、胚中蛋白亚基差异较小,只有个别亚基存在差异.

Abstract:对大花金挖耳内生真菌H-18菌株的菌丝提取物进行了抑菌活性研究并对菌株进行了鉴定.离体试验结果表明,H-18菌株菌丝体丙酮提取物在500mg/L浓度下对玉米大斑病菌、番茄灰霉病菌和辣椒疫霉病菌的菌丝生长抑制率均大于80%;组织法和盆栽试验结果表明,H-18菌株菌丝体丙酮提取物在2000mg/L浓度下对番茄灰霉病和辣椒疫霉病的保护效果和治疗效果均在50%以上.结合形态分类学和ITS-5.8S rDNA序列分析结果鉴定菌株H-18为同色镰孢菌(Fusarium concolor).

Abstract:采用营养液培养法研究了苗期龙葵(Solanumnigrum L.)对镉锌复合污染的反应及其对镉锌的吸收和积累特性.结果表明:(1)高浓度镉(200μmol·L-1)、锌(500μmol·L-1)胁迫使苗期龙葵植株生长受到严重抑制,但低浓度镉锌复合处理的部分植株生物量甚至超过了对照.(2)添加锌对苗期龙葵地上部镉积累有显著的影响(P<0.05);当锌浓度为100μmol·L-1时,镉积累量达到峰值,其中的100Cd/100Znμmol·L-1处理组合的镉积累量为所有处理中的最大值(地上部积累量高达171.31μg·plant-1);当锌浓度增加到500μmol·L-1时,植株体内镉积累量则呈现下降趋势.(3)添加镉对苗期龙葵地上部锌积累也有显著的影响(P<0.05);当镉浓度达50μmol·L-1时,根、茎、叶中锌积累量均达到最高值;当镉浓度为200μmol·L-1时,各处理单株地上部锌积累量最低.(4)从植株整体的镉、锌积累量来看,苗期龙葵地上部镉积累量最高达根系的14.67倍,而其锌积累量最高达根系的13.59倍.研究发现,龙葵幼苗根系、茎和叶中镉、锌含量随营养液中镉、锌浓度的增加而显著增加(P<0.05);添加锌在一定程度上缓解了镉对苗期龙葵的毒害效应,低浓度锌还可以提高植株对镉的吸收和积累;添加镉在一定程度上增强了植株对锌的耐受性和吸收富集能力;植株地上部的镉、锌积累量明显高于根系.

Abstract:以遮荫生长的盆栽西瓜幼苗为材料,研究了100mg/L5-氨基乙酰丙酸(5-aminolevulinic acid,ALA)处理对暗适应叶片转入强光下叶绿素荧光特性的影响.结果显示,遮荫能显著提高暗适应叶片的Fo,降低Fv/Fm和Fv/Fo;正常光照下生长的植株叶片暗适应后转入1500μmol·m-2·s-1作用光强下5min的ΦPSⅡ、qP和Pc分别为0.176、0.399和0.180,约为600μmol·m-2·s-1作用光强下的62%、72%和64%;而遮荫下生长的幼苗叶片暗适应后转入1500μmol·m-2·s-1作用光强下的ΦPSⅡ、qP和Pc分别为0.089、0.301和0.089,仅为600μmol·m-2·s-1作用光强的40%、66%和40%;遮荫还显著降低西瓜叶片暗适应后转入强光1500μmol·m-2·s-1的PCR和qL,同时提高L(PFD).ALA处理能提高遮荫西瓜幼苗叶片PCR、Pc、qL和Hd等荧光参数,降低Ex、ΦNO和L(PFD);SOD活性抑制剂DDC处理降低PCR、Pc、qL和Hd等荧光参数,而ALA处理可以逆转DDC的抑制效应;ALA处理能提高西瓜幼苗叶片SOD、POD和APX活性.研究发现,遮荫导致西瓜幼苗光抑制程度加重,ALA通过增强PSⅠ附近SOD等抗氧化酶活性,促进水-水循环,增加热耗散,减轻光抑制,提高西瓜幼苗叶片的光化学效率,从而对强光下的光合作用起到保护作用.

Abstract:以大穗小麦品种SN05-3和多穗小麦SN02-3为试验材料,研究了化学杂交剂BAU-9403对灌浆期小麦库、源、流的影响.结果表明,BAU-9403诱导的小麦雄性不育率大于95%,异交结实率高于92.5%;BAU-9403能够显著降低旗叶叶绿素含量和提高其净光合速率;喷药处理的干物质日生产量,茎、叶、鞘的干物质转移量及其输出率和转换率均下降;喷药处理籽粒饱满度平均减少0.222,平均灌浆速率降低0.306g/1000 grains·d-1,灌浆持续期平均缩短6.03d.BAU-9403处理的SN05-3净光合速率增加幅度、叶绿素含量及茎中干物质转移量、输出率和转换率下降幅度均高于SN02-3,而叶中干物质转移量下降幅度和鞘中的干物质输出率却低于SN02-3;SN05-3的灌浆持续期缩短幅度明显大于SN02-3,而其灌浆速率下降的程度小于SN02-3.综合分析表明,BAU-9403能够影响灌浆期小麦的光合生理特性,破坏库、源之间的平衡关系,使干物质积累和转运不畅,导致粒重下降,且品种间的反应存在差异.

Abstract:通过盆栽试验,研究了土壤水分含量对翅果油树幼苗地上部分和根系生物量和含水量,以及叶片保水力、相对电导率和保护酶活性的影响.结果表明:与对照(处理前)相比,各土壤水分条件下翅果油树幼苗叶片保水力均降低,而叶片相对电导率(REC)、超氧化物歧化酶(SOD)和过氧化物酶(POD)活性均升高.随着土壤含水量的降低,幼苗地上部分和根系的生物量、含水量及叶片保水力均呈先升高后降低的趋势,而叶片REC、SOD和POD活性均呈先降低后升高趋势,并均以轻度干旱胁迫处理(田间持水量的55%~60%)的REC、SOD和POD活性最低,而叶片保水力最高.在相同土壤水分条件下,随着处理时间的延长,叶片REC逐渐升高,而叶片保水力降低,SOD和POD活性先升后降并以处理10d时活性最高.研究发现,在轻度干旱胁迫下,翅果油树幼苗能协调其保护酶系统降低干旱伤害,维持正常生长,表现出一定的抗旱能力.

Abstract:在人工增强紫外线辐射条件下(UV辐射光波长308nm,辐射剂量18kJ·m-2·d-1),将浓度为1mg·L-1的芦荟蒽醌类物质喷施到菠菜叶表面(喷施量50mL·m-2),研究外源芦荟蒽醌类物质在增强UV辐射条件下对菠菜生长发育的影响.结果显示:在增强UV辐射条件下,与没有喷施蒽醌类物质的菠菜相比,喷施蒽醌类物质的菠菜株高增加5.40cm,叶面积增加18.66%,干鲜比增加22.28%,总叶绿素含量高出11.33%,POD和SOD活性分别增加了54.08%和21.05%,类黄酮含量则降低了28%.上述生理生化指标和不受紫外线辐射条件下生长的菠菜发育水平接近.实验结果表明,在植物体表面喷施外源芦荟蒽醌类物质能够有效减轻UV辐射对植物的伤害.

Abstract:以柠条、沙木蓼、杨柴和花棒4种沙生灌木幼苗为材料,采用盆栽法在适宜水分、中度干旱和重度干旱(田间持水量的75%、50%和35%)3种土壤水分条件下,应用PV技术测定了它们在膨压为0时的渗透势(ψstlp)、相对水含量(RWCtlp)和相对渗透水含量(ROWCtlp),以及饱和含水时的渗透势(ψssat)、束缚水含量(Va)、膨压随叶水势下降而降低的速率b值和组织细胞总体弹性模量(ε′)等水分参数,并用隶属函数值法对4种苗木在干旱下保持膨压的能力进行了综合评价.结果显示:与适宜水分条件下相比较,除花棒幼苗在中度及重度干旱下ψssat值、柠条苗在中度干旱下ψssat和ψstlp差值和在中度及重度干旱下Va值、沙木蓼苗在中度干旱下ε′和RWCtlp值均变化很小以外,4种苗木其它水分参数在不同程度干旱胁迫下均有较明显的变化,且其变化幅度随干旱胁迫的加剧而增加,从而使苗木保持膨压及吸水保水的能力较适宜水分下明显增强;苗木保持膨压能力的综合评价结果为:中度干旱下柠条>花棒>杨柴>沙木蓼,重度干旱下柠条>花棒>沙木蓼>杨柴.研究表明,柠条幼苗对干旱胁迫具有极强的渗透调节适应性.

Abstract:通过不同浓度外源谷氨酸(Glu)和γ-氨基丁酸(GABA)以及不同比例Glu/GABA处理梭梭种子,研究它们对梭梭种子萌发率、胚根长度和呼吸速率的影响.结果显示:(1)不同浓度的外源Glu均能显著提高梭梭种子的萌发率,促进胚根的伸长并增强其呼吸速率;(2)外源GABA在低浓度(0.1~5mmol·L-1)时能降低种子萌发率,促进胚根伸长,而高浓度(10mmol·L-1)的效应则相反;不同浓度GABA均抑制种子呼吸速率,且低浓度时抑制效果更明显;(3)不同比例Glu/GABA均能降低种子萌发率,抑制胚根伸长,增强呼吸速率.研究发现,梭梭种子的萌发和呼吸受到外源谷氨酸的促进,却受到外源γ-氨基丁酸的抑制.

Abstract:在不同施肥方式下研究了长期试验地夏玉米在4个典型生育期的叶绿素相对含量(SPAD)、叶面积指数(LAI)和冠层光谱特征,系统分析了单波段以及由可见光和近红外波段组成的归一化差异植被指数(NDVI)与叶片叶绿素含量和LAI的相关性.结果表明:在同一个生育期,NK、PK处理和CK的光谱曲线特征相似,NPK、NP、OM、1/2OM1/2NPK处理的光谱特征曲线相似.除460nm外,其它单波段反射率与叶绿素含量和LAI都显著相关,尤其在510~1100nm波段相关性最为密切;不同波段组合的NDVI与这2个指标也显著相关,且相关系数大于单波段.从拔节期到乳熟期,绝大部分NDVI都可以有效地拟合叶绿素含量和LAI的变化,选用NDVI(560,950)和NDVI(660,760)建立拟合模型(R2>0.60),并以NDVI(560,950)拟合效果最佳(R2>0.80).可见,不同施肥方式条件下长期试验地夏玉米冠层光谱特征与叶绿素含量、LAI都具有良好的相关性,可以为夏玉米肥料管理提供科学依据.
