Abstract:利用Genome walker法获得了荔枝营养贮藏蛋白质LcVSP1基因的5'调控序列,构建了含有该序列的植物表达载体并转化烟草,通过PCR和GUS染色对转化植株进行了鉴定.序列分析表明,LcVSP1基因的5'调控序列中除含有真核生物典型的核心启动子区域和保守的TATA box序列外,还发现了一些与真核生物启动子中相似的顺式调控元件.对获得的转基因植株的GUS染色发现,在转基因植株的茎、叶柄和主叶脉呈现蓝色,表明LeVSP1的5'调控序列可以启动GUS基因的表达,具有启动子的活性,并具有组织特异性.

Abstract:以油茶EST文库为基础,采用5-′RACE技术,分离克隆了一个脱水素样基因的全长cDNA序列(GenBank接受号EU856537),同源分析表明其编码的208 aa的小分子蛋白(id号ACF72673)属于SK2型脱水素,命名为CoDHN2.CoDHN2的肽链内2个类K-片段间富含苏氨酸,有别于脱水素一般性结构特点,并且同油茶种子中另一类脱水素一样也具有一个十分保守的基序(EDDGQAGRRKK),这可能有利于脱水素的磷酸化和亚细胞定位;采用多种方法预测其二级结构,表明CoDHN2为内在性无规则蛋白,但其中远离C端的一个类K-片段可形成两亲性α-螺旋.CoDHN2含有较多的组氨酸残基以及良好的可溶性,推测它可结合金属离子从而减少活性氧的来源并清除活性氧,并在生理脱水时充当缓冲液.结合其它物种脱水素的研究进展,认为CoDHN2极有可能在油茶油脂合成高峰期同正在发育的脂体结合而保护脂体免受活性氧危害,这为油茶种子细胞的脂体发育研究提出了一个新的方向.

Abstract:以豫麦18为受体导入反义trxs基因获得的株系01TY18(T0)为材料,运用PCR、实时荧光RT-PCR以及离体整穗发芽的方法,对反义trxs基因在转基因小麦中的遗传稳定性、基因表达和抗穗发芽特性进行了研究.结果表明,8个T1代转基因株系中有6个株系目的基因检测呈阳性,且在以后的世代中能够稳定遗传并呈典型的孟德尔单基因3∶1分离规律;反义trxs基因在6个转基因株系中能够正常表达且表现出显著的抗穗发芽特性.与非转基因对照相比,转基因株系穗粒发芽率和穗发芽度平均分别降低62%(P<0.01)和50.8%(P<0.01).

Abstract:采用秋水仙素染色体加倍技术,获得了纯合黑籽甘蓝型油菜品系NJ230(Brassica napus)的染色体加倍材料(2n=76).对加倍材料连续自交3代,观察加倍材料后代染色体数目及性状变异,结果表明:(1)加倍材料当代(D1)植株矮化;(2)加倍材料自交1代群体(D2)呈现雄性不育、种子颜色等性状变异;(3)加倍材料自交2代后(D3、D4),多数植株染色体数为2n≤38;(4)获得了1个2n=32的黄籽非整倍体系.本实验为油菜非整倍体创建提供了一种可行的方法.

Abstract:采用ITS序列分析和ISSR分子标记技术,对中国野生百里香(5个种2个变种)及国外百里香(2个种)共计20份材料进行了属内种间及种下水平的系统发育和亲缘关系研究.结果表明:(1)ITS序列的系统发育树能较好地说明百里香属种间亲缘关系,地椒、地椒亚洲变种、百里香和Thymus serpyllum共同聚为一组,Bootstrap支持率为96%,表现为较近的亲缘关系;(2)基于ITS序列的系统发育树和ISSR多态性标记的聚类分析均将地椒怀远居群和地椒展毛变种兴凯居群聚为单独的一组,表现出与其它供试材料间较为明显的遗传差异;(3)ISSR遗传多态性分析将地椒、地椒亚洲变种和百里香很好的聚类;种下居群间的聚类则体现了与地理分布之间一定的相关性.将本实验结果与野外考查及形态学比较相结合,可以得出以下结论:地椒、地椒亚洲变种和百里香三者之间属于近缘种,并与国外的T.serpyllum种间有很近的亲缘关系;支持《黑龙江植物志》将地椒展毛变种兴凯居群列为单独的一个种;地椒怀远居群在遗传背景及分布生境上有很大的特殊性,对其在百里香属植物中的系统地位仍需进一步研究确定.

Abstract:采用低剂量(5 mW·mm-2)He-Ne激光辐照经增强UV-B辐射处理(10.08 kJ·m-2·d-1)后的晋麦47号小麦幼苗,用常规石蜡切片法,观察各处理组小麦幼苗叶片组织显微结构特征;同时对幼苗光合特性、水分利用及色素含量状况进行测定分析.结果显示:(1)与对照(CK)相比,UV-B辐射处理(B)与复合处理(BL)的表皮细胞及腺毛排列致密,气孔稀疏、缩小且下陷;叶肉组织细胞均明显加厚,细胞略小且排列紧密,维管束相对面积减小,BL的以上性状均介于CK与B之间.(2)B组净光合速率、气孔导度、蒸腾速率均显著降低.与B相比,BL气孔导度及光合速率偏高,蒸腾速率差异不明显.就水分利用率而言,L最大,略高于CK,B组最小,略低于BL,各组间差异显著.(3)增强UV-B辐射可使小麦幼苗叶绿素含量显著降低,经He-Ne激光辐照处理后可以使叶绿素含量得到一定程度恢复性升高,并且Chla变化的幅度大于Chlb.结果表明:低剂量He-Ne激光辐照能够对小麦幼苗组织显微结构以及光合器官的UV-B辐射损伤起到促进修复的作用.

Abstract:采用液体培养研究Pb、NaCl单因子及复合胁迫对喜盐鸢尾(Iris halophilaPall.)的生长和生理变化的影响以及Pb在鸢尾植株不同部位的积累.结果显示,培养8 d后,随着Pb NaCl复合浓度的增加,喜盐鸢尾地上部和地下部干重下降;当Pb处理浓度提高,喜盐鸢尾地上部和地下部Pb含量显著增加,在5 mmol?L-1Pb 50mmol?L-1NaCl胁迫下,地上部Pb含量达到1 534μg?g-1,达到超富集植物的标准,但植株干重下降不显著;NaCl对Pb的吸收有一定促进作用.Pb、NaCl单因子或复合胁迫均增加了对喜盐鸢尾幼苗的伤害,在100 mmol?L-1NaCl及Pb和NaCl复合胁迫下,MDA含量和Pro含量显著增加,在10 mmol?L-1Pb、100 mmol?L-1NaCl及Pb和NaCl复合胁迫下SOD活性显著提高.结果表明,喜盐鸢尾具有一定的耐Pb和NaCl胁迫的能力,并对Pb具有潜在的超富集能力,是一种可用于修复Pb污染环境的Pb潜在超富集植物.

Abstract:在西双版纳热带雨林条件下测定了3种热带经济植物灯台树(Alstonia scholaris)、瓜拉那(Paullinia cu-pana)和海巴戟(Morinda citrifolia)雨季和雾凉季的光合作用、叶绿素荧光、抗氧化酶活性及膜脂过氧化等指标变化.结果表明:雨季灯台树、海巴戟和瓜拉那的最大净光合速率(Amax)分别为18.9、23.2和11.7μmol?m-2?s-1.雾凉季3种植物的Amax、气孔导度、羧化效率和光系统Ⅱ的最大光化学效率都比雨季显著降低,并以灯台树降低的幅度最小;3种植物的非光化学焠灭(NPQ)在雾凉季比雨季都升高,其中灯台树的NPQ上升的幅度最大.雾凉季低温还导致了3种植物的抗氧化酶(CAT、SOD、APX、POD和GR)活性和丙二醛(MDA)含量都升高,并以灯台树抗氧化酶的活性更强或上升幅度最大,而MDA含量增加幅度最小.研究发现,与灯台树相比,海巴戟和瓜拉那对西双版纳雾凉季低温很敏感,雾凉季时其光合系统都受到一定程度的伤害,需要采取适当的防寒措施以缓解低温伤害.
